Abstract
The objective of this study was to develop multi-layer nanoemulsions (NEs) using a layer-by-layer technique to improve the cellular uptake and intragastric stability of α-Tocopherol (α-TC). α-TC-loaded multi-layer NEs were prepared using lecithin (PRI-NE) followed by coating with chitosan as the second layer (SEC-NE) and dextran sulfate as the third layer (TER-NE) using a high-energy-method. Multi-layer NEs were ranged from 96 to 160 nm and exhibited a high entrapment efficiency of 80-91%. Cellular association, antioxidant activity, and in vitro bioaccessibility of α-TC-loaded multi-layer NEs were significantly better than those of non-nanoencapsulated α-TC (p<0.05). The cellular association of TER-NE was the highest, 1.3-2-fold higher than that of non-nanoencapsulated α-TC. Also, the cellular antioxidant ability was PRI-NE (40%), SEC-NE (45%), and TER-NE (50%). The amount of free fatty acids released after digestion was decreased after multi-layer nano-encapsulation. Moreover, bioaccessibility of TER-NE was two-fold higher than that of non-nanoencapsulated α-TC. These results indicated that the cellular association, antioxidant activity, gastrointestinal stability, and in vitro bioaccessibility of α-TC could be enhanced by multi-layer nano-encapsulation. Furthermore, TER-NE was the most effective multi-layer NE structure to improve cellular association and intragastric stability.
