Abstract
전압의존성 칼슘채널은 효소 활동, 신경전달물질 분비, 뉴런의 흥분성, 신경돌기의 성장 및 유전자 전사등과 같이 생화학적 작용에 전기적 신호를 전달하는 중요한 경로이다. 이러한 칼슘채널은 크게 2가지로 고전압의존 (HVA)과 저전압의존 (LVA or T-type)으로 분류된다. 특히, T-타입 칼슘채널은 현재 고혈압, 간질 그리고 신경성 통증을 치료할 수 있는 새로운 표적으로 주목받고 있다. 이를 위해 구조기반 (Structure Based Drug Design) 가상검색 (Virtual Screening) 기술을 이용하여 N-phenylsuccinimide 구조를 갖는 화합물들이 T-타입 칼슘채널에 저해작용을 하는 것을 알아냈다. 제안된 구조를 최적화하기 위해 T-타입 칼슘채널 저해제에 선택성을 갖는 화합물들과의 약리단을 조사하였고 40여개의 유도체들을 합성하여 α_(1G) 와 α_(1H) in vitro 검색 시스템을 통해 T-타입 칼슘채널에 대한 억제 활동에 대해 시험하였다. 그 결과, 8개의 N-phenylsuccinimide 유도체들이 Mibefradil 만큼 효과적인 in vitro 결과를 보여주었다.; Voltage-dependent calcium channels are the primary routes for translating electrical signals into biochemical events underlying key processes such as enzyme activity, neurotransmitter release, neuronal excitability, neurite outgrowth and gene transcription. Calcium channels have been subdivided into two major classes: high-voltage activated (HVA) and low-voltage activated (LVA or T-type) calcium channels. T-type calcium channels are now attracting much attention as novel therapeutic targets such as hypertension, epilepsy and neuropathic pain. We found several new hit compounds for T-type Ca^(2+)channel inhibitors based on SBDD (Structure Based Drug Design). Among them, the scaffolds to have N-phenylsuccinimide had the most potent activity against T-type Ca^(2+) channel 0.10 μM. We designed new compounds by pharmacophore mapping with N-phenylsuccinimide compounds in order to improve the activity and to know the structure activity relationship and we constructed the small library of N-phenylsuccinimide. Some of the synthesized compounds showed as good in vitro inhibitory activities as Mibefradil.
