키랄 아지리딘으로부터 순수한 부분입체 이성질체 1,4-디데옥시-1,4-이미노-D-아라비니톨의 전합성

Abstract

1,4-Dideoxy-1,4-imino-D-arabinitol은 Angylocalyx boutiqueanu에서 1985년에 분리되었으며, α-glucosidase 저해효과를 갖는 것으로 알려져 있다. α-glucosidase 저해제는 carbohydrate metabolism의 저해 기전에 의한 당뇨병 치료제로서 시장에 출시되어 있다. 그리고 항암제, 항바이러스제, 세포사멸유도체 등에도 적용 가능하여 관심을 받고 있으며, 이에 대한 연구/개발이 활발히 진행되고 있다. 1,4-Dideoxy-1,4- imino-L-arabinitol은 α-glucosidase 저해효과는 D-형에 비해 미미하지만 HIV 저해 효과가 우수하다고 알려져있다. 본 연구에서 최초로 D-arabinitol, L-arabinitol과 이들의 stereoisomer인 D-xylitol, L-xylitol을 상업적으로 얻을 수 있는 chiral aziridine 2-methanol로부터 총 5단계에 걸쳐 광학적으로 순수하고 고효율적으로 합성을 완료하였다. 그리고 이 합성법에서 가장 중요한 stereoisomer의 분리와 정제를 에탄올을 이용한 재결정을 통해 쉽게 할 수 있었다.; 1,4-Dideoxy-1,4-imino-D-arabinitol first isolated from the fruits of Angylocalyx boutiqueanus in 1985 turned out to possesses α-glucosidase inhibitory activity. The α-glucosidase inhibitors have proved to block carbohydrate metabolismand been successfully launched on the drug market for diabetes treatment. Moreover, it has also been reported α-glucosidase inhibitors exhibit antitumor, antiviral and apoptosis-inducing activities. The broad spectrum of biological activity of α-glucosidase inhibitors led us to be interested in synthesizing 1,4-dideoxy-1,4-imino-D-arabinitol, α-glucosidase inhibitor in highly efficient and stereoselective fashion. We would like to report a chiral aziridine could be readily transformed into 1,4-dideoxy-1,4-imino-D-arabinitol in chirospecific manner via ring opening of chiral aziridine, followed by ring formation of five-membered imino sugar.
