Abstract
Nanotechnology has been developed as a major innovative scientific and economic growth area around the world. However, manufactured nanomaterial may have potential hazardous effects on the marine ecosystem. To uncover the effect of nanomaterial on the mortality and antioxidant defense system in the rotifer, Brachionus sp., we analyzed the survival rate after exposure to different nanomaterials (eg., metal oxide nanoparticles (CuO, ZnO), and silver nanoparticle), carbon nanotubes (CNT), and fullerene (C60). We also cloned the cDNAs encoding oxidative stress-related genes from the rotifer genome database, and analyzed specific expression of these genes following exposure to nanomaterials using quantitative real-time RT-PCR. Through standard acute toxicity test procedures, several toxicity endpoints such as LC50, LC10, and no observed effective concentration (NOEC) of silver-nanoparticle were obtained. The oxidative stress-related genes chosen here were glutathione S-transferase (GSTs) family (alpha, omega, sigma, theta, and zeta), catalase (CAT), and manganese superoxide dismutase (MnSOD). After exposure to different concentration of nonamaterials, all the oxidative stress-related genes were significantly up-regulated with time course manner. The significant high levels of expression on the oxidative stress genes would suggest that the nanoparticles involved in occurring oxidative damage. These results support the hypothesis that nanoparticle induces oxidative stress in this species, and can be useful to test the effect of those chemicals on this organism.
