Abstract
Dip-pen nanolithography (DPN) and polymer pen lithography (PPL) are methods for generating biomolecule patterns because those are relatively simple and cost-effective techniques. DPN can fabricate various patterns with nanoscale resolution but is limited in throughput. PPL was developed to solve the limitation of throughput. However, it has several restrictions such as slow transportation of biomolecules from tips to a substrate, difficulty of preventing denaturation of biomolecules at the tip surface, short operation time due to the limited ink volume. Several surface modification methods and additives of both polyethyleneglycol and glycerol have been used to solve these problems. Agarose gel is well-characterized, hydrophilic, biocompatible, and water absorbable polymer. This characteristics make it possible to store inking solution in porous structure of the agarose gel tip array and, thus, to perform large-area pattern of biomolecules and to prevent drying biomolecular inks without adding carrier materials. In this study, the agarose gel was utilized as a tip materialfor fabricating large area biomolecule patterns. The Fluorescein isothiocyanate-labeled streptavidin (FITC- streptavidin) patterns were fabricated by the stamping cycles on a silicon substrate.Fluorescence intensity and AFM topography were used to confirm uniformity of patterned FITC- streptavidin dots and to measure size of dots, respectively. This thesisintroduces a simple and reproducible technique to fabricate biomolecule pattern without any surface modification methods and additives. This hydrogel–based PPL is suitable for fabrication of biomolecule pattern and it is potentially useful in biological fields.
