Abstract
Medium osmolality is an important factor for cell growth in mammalian cell cultures. The cell’s internal osmolality is typically 270-300mOsm/kg, which is the same range as the general medium osmolality. When a medium osmolality increases substantially, the cell growth is detrimental. We have investigated the effects of medium osmolality on cell growth and antibody production using a recombinant Chinese Hamster Ovary DG44 Cell (rCHO-DG44) and a chemically-defined medium, which was developed in our laboratory. Medium osmolality was adjusted by adding NaCl at various concentrations. In batch cultures, when the medium osmolality was higher than 455mOsm/kg and lower than 205mOsm/kg, the cell growth rate and antibody productivity started to decrease. When the osmolality was 255-355mOsm/kg, the cell achieved the highest growth and antibody productivity. 243mOsm was the lowest osmolality that does not cause severe impediment to the cell growth and antibody production. In the fed batch culture, Cell line 1 and 2 both showed effect of longevity and high antibody production at 255mOsm/kg. Cell line 1 showed these effects at 230mOms/kg, too. Death of cells was observed in the Cell line 1 when it was above 305mOsm/kg. It is because as osmolality rises, aspartic acid’s consumption rate gets faster and it causes depletion. As osmolality rises, amino acid is depleted and viability reduces. Therefore, fed batch culture at 230 and 255mOsm/kg is effective in producing antibody by preventing depletion of particular amino acid and reduction of viability. It can be utilized for fed batch culture.
