- A lateral flow immunoassay (LFIA) is widely used in clinical diagnostics due to its simplicity, rapidity, and cost-effectiveness. However, it has limitations in terms of detection sensitivity and quantification capabilities. In this study, the surface-enhanced Raman scattering (SERS)-based LFIA with high detection sensitivity is introduced for determining thyroid-stimulating hormone (TSH) concentration in clinical fluids. In this new LFIA biosensor, Raman reporter-labeled gold nanoparticles (AuNPs) are used as SERS detection probes. With the LFIA test strip, the presence of TSH can be first identified through a color change in test line, and then the quantitative analysis is performed by measuring SERS signals. The limit of detection (LOD) was estimated to be 0.025 μIU/mL, which is approximately 60 orders-of-magnitude more sensitive than that achieved with the conventional POC-based LFIA method. It is demonstrated that combining SERS with LFIA is beneficial due to its high detection sensitivity that is comparable to other conventional methods. Therefore, this novel SERS-based LFIA method has a strong potential feasibility which can be applied to the immunoassays of many other biomarkers in clinical diagnostics.
