- An amphipathic molecule has a tendency to self-assemble into nanostructures in an aqueous medium such as micelles and liposomes. Micellar nano-assemblies usually have the ability to encapsulate hydrophobic guest molecules and deliver water-insoluble drugs into target cells. Also, micellar self-assemblies are widely used to maintain membrane protein stable in an aqueous environment. Another typical self-assembled nanostructure is liposome with bilayer structure. This nanostructure contains the hydrophilic interior and thus can be utilized to load water-soluble drug molecules. The morphology and size of a self-assembled nanostructure are critical for its functional application. Thus, it is of great interest to know about how selfassembled nanostructures are engineered by structural modifications of building blocks. In this context, we have designed and prepared two classes of novel amphiphiles with glucose head groups. One class was derived from benzene, designated as benzene-derived facial amphiphiles (BFAs) and the other class is malonate-based agents, designated as tandem malonate-derived glucosides (TMGs). BFAs were applied for membrane protein study while TMGs were used to investigate structural factors of building blocks for engineering self-assembled nanostructures. The study with BFAs indicates that the new facial amphiphiles have favorable property than conventional detergents for membrane protein stability. In the second part of study using TMG architecture, we found that self-assembled nanostructures could be engineered by small structural variation in alkyl chain length of the liphophilc groups or the chain length of the spacers between two malonate units.
