- Obesity is now defined by the World Health Organization (WHO) as a disease as a disease causing various adult diseases. The World Obesity Federation (WOF) estimates if the current trend of obesity increases, one third of the world's population in 2025 will become obese (body mass index over 30kg/㎡). Obesity induces metabolic syndromes such as diabetes, hypertension, hyperlipidemia, cardiovascular disease, and the risk of death is as high as 30 %. In addition, the negative social prejudice leads to depression and stress, which in turn affects mental and daily life.
To treat obesity and obesity-related diseases emerging as social problems, much research has been done on adipocytes-specific delivery. Currently, studies on the treatment of obesity through gene delivery have been actively conducted.
The aim of this research is to overcome obesity through protein drug delivery with less risk of side effects. Protein transduction domain (PTD) fused with a short peptide, prohibit targeting sequence (PTS) enhances the cellular permeability and targetable ability of the protein to adipocytes. Prohibitin targeting sequence (PTS) binds to prohibitin located on the adipocyte surface and mitochondria enhancing the retention of protein into adipocytes. This was confirmed by cell based experiments and obese mice. The recombinant targetable fusion protein delivery system will contribute to the treatment of obesity and obesity induced metabolic syndromes.
