dc.description.abstractAsteadysupplyofplateletsmaintainstheirlevelsintheblood,andthisisachievedbythegenerationofprogenyfromplateletintermediates.Usingsystematicandintegrativesuper-resolutionmicroscopy,weexaminedtheultrastructuralorganizationofvariousorganellesindifferentplateletintermediatestounderstandthemechanismoforganelleredistributionandsorting,whichmayberequiredforplateletproduction.Weobservedthedynamicinterconversionbetweentheintermediatesviavariouspathwaysandfoundthatmicrotubuleswereresponsibleforcontrollingtheoverallshapeofplateletintermediates.Super-resolutionimagesshowedthatmostoftheorganelleswerelocatednearthecellperipheryinovalpreplateletsandconfinedtothebulboustipsinproplatelets.Wealsofoundthatthedistributionofthedensetubularsystemandα-granuleswasregulatedbyactin,whereasthatofmitochondriaanddensegranuleswasgovernedbymicrotubules.Altogether,ourresultscallforareassessmentoforganelleredistributioninplateletintermediates.-
