dc.description.abstract | Asteadysupplyofplateletsmaintainstheirlevelsintheblood,andthisisachievedbythegenerationofprogenyfromplateletintermediates.Usingsystematicandintegrativesuper-resolutionmicroscopy,weexaminedtheultrastructuralorganizationofvariousorganellesindifferentplateletintermediatestounderstandthemechanismoforganelleredistributionandsorting,whichmayberequiredforplateletproduction.Weobservedthedynamicinterconversionbetweentheintermediatesviavariouspathwaysandfoundthatmicrotubuleswereresponsibleforcontrollingtheoverallshapeofplateletintermediates.Super-resolutionimagesshowedthatmostoftheorganelleswerelocatednearthecellperipheryinovalpreplateletsandconfinedtothebulboustipsinproplatelets.Wealsofoundthatthedistributionofthedensetubularsystemandα-granuleswasregulatedbyactin,whereasthatofmitochondriaanddensegranuleswasgovernedbymicrotubules.Altogether,ourresultscallforareassessmentoforganelleredistributioninplateletintermediates. | - |
