dc.description.abstract혈소판활성화분자메커니즘을이해하는것은혈소판활성화상태를확인하고기존의지혈장애치료법을개선하는데필수적이다.본연구에서는,파라메톡시암페타민(PMA)자극으로인한활성화과정동안혈소판의시공간적초미세구조변화를다양한최첨단초고해상도이미징을통해관찰하고이를정량화하였다.초고해상도이미지를통해혈소판이액틴(actin)과미세소관(microtubule)의도움으로퍼질수있으며,대부분의세포기관은스펙트린(spectrin)-및비멘틴(vimentin)-막(membrane)골격의도움으로중앙에집중화될수있음이관찰되었다.또한,중앙집중화된세포기관중과립들이OCS와융합되어,이를통해그내부물질들을방출할수있음을관찰하였다.이러한연구결과를바탕으로지금까지보고되지않은PMA에의해유도된혈소판의활성화과정에대해다양한세포소기관의나노스케일적변화를관찰및정량화함으로써복잡한혈소판활성화과정의새로운패러다임을제공할수있었다.-
