- 남극 해양 규조인 Chaetoceros neogracile 은 저온 환경에 적응하기 위해 antifreeze protein (AFP) 합성하여 세포 외로 분비한다. 우리는 C. neogracile 에서 분비되는 AFP (Cn-AFP)를 암호화 하는 유전자를 클로닝 하여 이 유전자에 대한 특성을 연구하였다. Cn-AFP cDNA 의 길이는 994 base pair 이며 이 중 open reading frame (ORF) 은 849 base pair 이다. Cn-AFP ORF 는 282 amino acid 로 이루어져 있으며 약 31 kDa 의 분자량을 갖는다. 이 중 N-terminal 의 30 개 amino acid 는 signal peptide 로 밝혀졌으며 이 signal peptide 에는 glycosylation site 와 myristoylation site 가 존재한다. Signal peptide 가 제거된 mature Cn-AFP 는 약 27 kDa 의 분자량을 갖고 있다.
Cn-AFP 의 copy number를 알아보기 위해 genomic Southern Blot Analysis 수행하였다. 이 결과를 통해 Cn-AFP 는 C. neogracile 내에 multi-gene family 로 존재한다는 것을 확인할 수 있었다. 그리고 freezing stress 조건에서 Cn-AFP 유전자의 발현이 급격히 증가한다는 것을 Northern Blot Analysis 결과로 알 수 있었다. 같은 조건 하에서의 protein 의 발현 변화와 세포 외로 분비되는 것을 확인하기 위해 세포를 배양한 배양액을 여과, 농축하여 Western Blot Analysis 를 수행하였다. 그 결과, Cn-AFP 는 세포 외로 분비된다는 사실과 freezing stress 조건 하에서 Cn-AFP protein 발현이 증가함을 알 수 있었다.
Cn-AFP 의 대량 발현과 antifreeze activity 를 측정하기 위해 pre-mature Cn-AFP 와 mature Cn-AFP 를 protein expression vector 인 pProEX HTa 에 삽입한 후 E. coli BL21(DE3)에 transformation 하여 과발현 시켜 정제하였다. 이렇게 정제된 recombinant pre-mature, mature Cn-AFP 와 C. neogracile 에서 분비된 AFP 는 다양한 antifreeze activity 을 나타내었다. Recombinant mature Cn-AFP 는 thermal hysteresis (TH) activity 를 보였으며 최고값이 0.4 ℃ 이다. 또한 recombinant pre-mature, mature Cn-AFP 모두 morphological change of ice crystal activity 와 ice-pitting activity 을 나타냈다. 하지만 mature Cn-AFP 가 pre-mature Cn-AFP 보다 더 좋은 활성을 보였다. 또한 세포에서 분비된 Cn-AFP 도 활성을 나타내었지만 recombinant Cn-AFP 보다 낮은 활성을 보였다.; The Antarctic marine diatom, Chaetoceros neogracile produced an antifreeze protein (AFP) and secreted it to the extracellular space. The gene of the antifreeze protein (Cn-AFP) from the Antarctic diatom, C. neogracile, was cloned and characterized. The Cn-AFP cDNA clone contained a full-length open reading frame of 849 bp and the deduced 282 amino acid peptide chain encodes a 31 kDa protein, which includes a signal peptide of 30 amino acids at the N-terminus. The signal peptide included a glycosylation and myristoylation site. The molecular mass of a mature Cn-AFP without signal peptides is 27 kDa. The Cn-AFP is encoded by a multi-gene family that was investigated by Southern Blot Analysis. Northern Blots demonstrated that the Cn-AFP transcript was dramatically accumulated when C. neogracile was treated with freezing stress. Western Blot Analysis revealed that the level of secreted Cn-AFP was also induced by freezing stress. The full-length Cn-AFP genes with pre-mature Cn-AFP and mature Cn-AFP were cloned into an E. coli expression vector and expressed as a fusion protein. The native Cn-AFP, purified recombinant pre-mature and mature Cn-AFP showed various degrees of specific antifreeze activity. Recombinant mature Cn-AFP has thermal hysteresis (TH) activity. The highest value of recombinant mature Cn-AFP TH activity is 0.4°C, and recombinant pre-mature and mature Cn-AFP show a morphological change of ice crystal activity and ice-pitting activity. However, recombinant mature Cn-AFP has more morphological change of ice crystal activity and ice-pitting activity than recombinant pre-mature Cn-AFP.
