dc.description.abstractThe objective of this research was to evaluate the transfection efficacy of the reducible poly (oligo-D-arginine) (rPOA) to enhance the therapeutic efficacy of ganciclovir/herpes simplex virus thymidine kinase (GCV/HSV-tk) suicide gene therapy for brain tumor. As a result of in vitro stability, cytotoxicity, and transfection efficiency of DNA/ rPOA polyplex, rPOA is a promising non-viral gene delivery carrier and protein transduction domains (PTDs), providing lower cytotoxicity and similar gene expression efficiency compare to PEI. In addition, brain tumor cells were showed high cytotoxicity by rPOA/HSV-TK gene expression in comparison with PEI in the presence of ganciclovir (GCV). And the next step, the ability of the rPOA to deliver the HSV-tk confirmed that functional testing of hind limb movement was assessed using the Basso, Beattie, and Bresnahan (BBB) scoring rubric. The results demonstrated that the rPOA/HSV-tk could transfect to tumors and anti-tumor efficacy was significantly higher than HSV-tk without rPOA in the presence of GCV.-
