dc.description.abstractCells secrete many extracellular vesicles such as micro vesicles, exosomes and membrane particles. It has been studied that exosomes had characteristics of their cell’s characteristics and normally, exosomes have diameter from 30nm to 100nm. In this study, we used human Mesenchymal Stem Cells (hMSCs) derived exosomes to treat Myocardial Infarction. For synergistic effect, Metallothionein protein was added which was transfected to exosomes. Briefly, Tat-metallothionein recombinant proteins were constructed. Tat (Protein transduction domain) was added to improve penetration of metallothionein in to hMSCs derived exosomes. Exosomes transfected by Tat-metallothionein recombinant proteins were purified. Metallothionein fusion protein of exosomes were studied by Western blot and visualized through confocal laser scanning microscopy (CLSM). In hypoxic conditions, exosomes with metallothioneins showed more therapeutic effects than exosomes. By this study, it can be suggested that exosomes are good carriers for proteins and have therapeutic effects in hypoxic conditions.-
