Abstract
Reactive oxygen species (ROS) are endlessly generated and eliminated in the diverse biological system and are required to induce regulatory pathways. Excessive ROS can damage lipids, cellular proteins and DNA under oxidative conditions. Therefore, we have synthesized water soluble selenium compounds that can scavenge ROS. The selenium compounds were removed excess produced ROS during the oxidation process. This experiment confirmed the ROS scavenge effect of selenium compounds in H2O2 environment using UV-spectrometer, 1H-NMR and FT-IR. UV-spectrometer results confirmed the reduction of selenide bond at 303nm. Result of 1H-NMR was shown about chemical shift by destruction of selenide bond. According to FT-IR, the selenic acid formed by the destruction of the selenide bond was confirmed at 997cm-1. In addition, β-chitosan based nanoparticles were prepared with the selenium compounds. Dissociation effect of nanoparticles by ROS was confirmed using DLS and TEM. As the concentration of H2O2 increases, the size of the nanoparticles decreases. The safety of the nanoparticles in normal cells were confirmed by FACs and CCK-8 assay. According to FACs and CCK-8 assay datas Chi-diSe-HyA-TPP NPs were safety up to 200 ㎍/ml.
