Abstract
The aims of this study were to optimize the extraction conditions of Kudzu root and to develop a Kudzu root extract?loaded nanoparticle for improving its water solubility. For this purpose, response surface methodology was used to optimize the extraction conditions, including ethanol concentration (X1) and extraction temperature (X2) to obtain the maximal total isoflavone and puerarin content in Kudzu root extract. Then we prepared Kudzu root extract-loaded nanocapsules using chitosan and poly-?-glutamic acid (?-PGA) and investigated the effect of Kudzu root extract concentration on the particle characteristics, such as particle size, polydispersity index (PDI), zeta potential, entrapment and loading efficiency. In addition, the effect of nanoencapsulation on the solubility of Kudzu extract was also evaluated. The optimized values of X1 and X2 were found to be 95% and 70?C, respectively. Experimental results obtained for the optimum extraction conditions agreed favorably with the predicted results, indicating the usefulness of prediction models for obtaining the maximal total isoflavone and puerarin content in Kudzu root extract. As the initial concentration of Kudzu root extract was increased, the nanoparticle size and loading efficiency were significantly increased. Nanoencapsulation of Kudzu root extract using chitosan and ?-PGA improved its solubility. These results suggest that the physical entrapment of Kudzu root extract within chitosan/?-PGA matrix could be useful for improving the solubility of Kudzu root extract.
