Abstract
Cancer is one of major concerns in human public health in the world. Chemotherapy is a one of widely adapted methods for treatment of cancer. However, it is often limited due to the side effect of anti-cancer drugs. Therefore, various drug carriers have been developed to overcome the limitation. In this thesis, alginate-based, thermally reversible, self-assembled polymeric micelles were designed and suggested for anti-cancer drug delivery. Alginate-g-poly(N-isopropylacrylamide) (PNIPAAm) was synthesized by conjugation of amino-PNIPAAm to the alginate chain via carbodiimide chemistry. Alginate-g-PNIPAAm formed micelles at the body temperature and the size of the micelles was varied depending on the molecular weight of PNIPAAm, degree of substitution, and polymer concentration. Doxorubicin (DOX), used as a model anti-cancer drug, was efficiently encapsulated in the micelles, and the sustained release of DOX from the micelles was achieved in vitro. These micelles were accumulated at the tumor site of a mouse model by enhanced permeability and retention effect. Interestingly, DOX-loaded alginate-g-PNIPAAm micelles showed enhanced anti-cancer efficacy in a tumor-bearing mouse model compared with soluble DOX alone.
