Abstract
Stimuli-responsive polymeric systems have been widely used for many drug delivery and tissue engineering applications. Magnetic stimulation can be also used to control the release of pharmaceutical drugs, growth factors and cells on-demand (on/off). To modulate the drug release, alginate-based ferrogels containing iron oxide nanoparticles were fabricated via ionic cross-linking. In general, the release of water-soluble drug from hydrogels is fast and difficult to achieve the sustained release for a long time period. In this thesis, the release rate of drug loaded in ferrogels was first regulated either by physically adding gelatin particles or by chemically introducing heparin to the gels. Gelatin particles were fabricated via a water-in-oil emulsion method, which delayed the release of trypan blue, a model water-soluble drug, from ferrogel, and the release rate was substantially regulated by applying the magnetic field. Alginate-g-heparin was also synthesized via carbodiimide chemistry. TGF-beta 1 was bound to heparin and used as a model growth factor. The sustained release of TGF-beta 1 from alginate-g-heparin gels was achieved and its release was regulated on-demand under the magnetic field. Alginate-based ferrogels that release drugs in a controlled manner may be useful in many applications including drug delivery and tissue engineering areas.
