dc.description.abstracthTMSC) 스페로이드 복합체에서도 높은 증식과 생존능을 보였음. 더욱이 세포와 섬유체는 스페로이드 복합체 내부에서 고르게 분포되어 있었고, TEM 분석을 통해 세포가 세포 간 결합을 유지한 상태로 PD-FF에 단단히 고정되어 있음을 확인함. 이러한 특성을 통해 줄기 세포의 성장인자 분비와 stemness가 높게 유지되었으며 분화 배지에서는 향상된 골 분화능을 확인할 수 있었음. 또한 줄기 세포의 기능은 FF의 미네랄화를 통해 조절 가능했음 (mFF). 미네랄은 FF 표면에 고르게 코팅 가능했음. Alizarin 염색을 통해 인간 지방 유래 줄기세포 (human adipose-derived stem cell; hADSC) 스페로이드 복합체 내부에 mFF가 고르게 분포함을 확인함. 또한 mFF는 골 분화 마커의 발현을 증가시켰음. 이후 스페로이드 복합체의 융합을 통해 밀리미터 단위의 조직체를 제작하는 것이 가능했음. 미네랄화된 스페로이드 복합체는 하나의 골 유사 조직으로 융합되었으며, 세포와 미네랄의 고른 분포를 보였음. 끝으로 골 조직 내부의 신호 전달 체계를 모사하기 위해, FF는 아데노신-폴리도파민 코팅을 통해 표면 개질되었음 (AP-FF). 폴리도파민은 섬유체 표면의 아데노신 코팅을 안정적으로 유지할 수 있게 도와주었음. 또한 AP-FF가 포함된 hADSC 스페로이드 복합체에서 향상된 아데노신 2b 수용체의 (a2bR) 발현을 확인할 수 있었음. 이러한 a2bR 신호 체계의 활성화는 일반 배지에서의 골 분화 마커의 향상된 발현을 유도했을 뿐만 아니라 분화 배지에서는 시너지 효과를 보이는 것으로 확인되었음. 더욱이 a2bR 신호 체계의 활성화는 지방 분화 배지에서 hADSC의 지방 분화를 크게 저해하는 것으로 확인되었음. 그리고 스페로이드 복합체의 피하 이식을 통해서도 생체 내 향상된 미네랄 증착과 골 분화 마커 발현을 확인할 수 있었음. 마지막으로 이 스페로이드 복합체의 골 결손 모델로의 이식을 통해 향상된 골 조직 재생능을 확인할 수 있었음. 따라서 이와 같은 스페로이드 복합체는 생체 내 세포 이식을 위한 전달체, 체외 조직 모델의 제작, 3D 바이오 프린팅과 같은 여러 응용법에 활용될 수 있을 것으로 예상됨.; There is a high demand for the functional bone grafts for the treatment of diseases associated with the bone such as trauma, injury, and degenerative diseases including osteoarthritis and osteoporosis. For example, more than one million patients receiving bone grafts annually in the United State alone, at a cost greater than $5 billion. In the last few decades, tissue engineering approaches have been proposed as an alternative therapeutic strategy for the treatment of bone diseases. Conventionally, tissue engineering approaches utilized stem cell, a scaffold for 3 dimensional (3D) support for cell attachment, growth, and proliferation, and bio-signaling molecules to stimulate the differentiation of stem cells. However, still, these approaches remained challenging to design the suitable scaffold to fulfill the needs of fields. With the recent development in understanding the intricate interactions of the cell with their surrounding microenvironment, now researchers are focused to fabricate tissue engineering scaffolds by mimicking the structural and compositional cues of extracellular matrix (ECM). Interstingly, electrospining is a cost effective process to fabricate nanofiber with a wide range of materials. Therefore, electrospun nanofiber has been widely explored in tissue engineering due to their structural similarities with ECM, providing large surface area and porosity which favors the cell migration, attachment, and proliferations. In addition, nanofibers are further surface modified with bioactive molecules to better control the differentiation of cells. However, nanofibers are collected in 2D sheet form and still, has limitation to fabricate 3D tissue. Here to overcome these limitations, I developed a composite spheroid incorporating engineered ECM prepared from fragmented electrospun fibers to tune stem cell functions. Firstly, an electrospun fibrous mesh of poly (ι-lactic acid) (PLLA) was aminolyzed into fragmented fibers (FFs), subsequently polydopamine coating for cell attachment. For preliminary characterization composite of fibroblast were fabricated which showed a significant increase of DNA content and viable nuclei in hybrid-spheroids. Similarly, composite showed significantly higher proliferation and viability of human turbinate mesenchymal stem cells (hTMSCs). Moreover, cells and fibers were homogenously distributed in composite spheroids, TEM analysis confirmed the cells tightly bound to PD-FF as well as maintaining cell-cell contacts. These characteristics led to significant growth factor secretion, preservation of stemness under the growth media, and osteogenic differentiation upon induction. Then, stem cell functions were controlled by a mineralized coating of FFs (mFF). Minerals were homogeneously coated on FFs surface. Alizarin staining revealed the homogenous distribution of mFFs in composite spheroids of human adipose-derived stem cells (hADSCs). Further, mFFs incorporation upregulated the expression of osteogenic markers. Then, a millimeter-scaled tissue construct was fabricated with a fusion of composite spheroids. The mineralized composite spheroids were compactly fused into single bone-like tissue construct revealing the homogenous distribution of cells and minerals. Finally, to mimic the bone signaling microenvironment, the FFs were modified with one-step adenosine polydopamine coating named AP-FF to induce adenosine signaling in composite spheroids for osteogenic differentiation. The polydopamine significantly enhanced the adenosine coating and retention on the fiber surface. Moreover, AP-FF incorporation into hADSCs composite spheroids significantly enhanced the adenosine 2b receptor (a2bR) expression. This activation of A2bR signaling led to significant upregulation of osteogenic markers in growth media and the synergetic effect was observed in osteogenic media. Moreover, A2bR signaling significantly inhibited the adipogenic differentiation of hADSCs in adipogenic differentiation media. Then, implantation of composite spheroids in subcutaneous tissue also showed significant minerals deposition and osteogenic markers expression in the ectopic region. Finally, implantation of composite spheroids in mouse calvarial model significantly enhanced the bone regeneration. Taken together, this composite spheroid platform can be an alternative approach for in vivo cell delivery carrier, in vitro tissue model development, and for bioprinting applications.; 외상 및 골다공증, 관절염과 같은 퇴행성 질환의 치료를 위한 기능성 골 이식제에 대한 수요가 증가하고 있음. 미국에서는 매년 100만명 이상의 환자가 골 이식술을 받고 있으며 그 비용은 50억 달러 이상임. 따라서 지난 몇 년 동안 골 질환 치료를 위한 대체 치료 전략으로 조직 공학적 접근법이 연구되었음. 조직 공학적 접근은 줄기 세포와 세포 부착 및 성장 유도, 생체 신호 전달을 위한 3차원 지지체를 활용하였음. 그러나 이러한 접근법으로는 효과적인 골 이식체 제작이 어려웠음. 이를 해결하기 위해 최근 세포와 그 주변 환경의 복잡한 상호 작용을 이해하고, 세포 외 기질의 구조를 모방하여 골 조직 재생에 필요한 지지체를 제작하는 연구가 진행되고 있음. 그 한가지 방법으로서 전기 방사는 세포 외 기질을 모방한 나노 섬유를 제작하기 위한 효율적인 방법으로, 제작된 나노 섬유는 세포의 분화를 제어하기 위해 생체 활성 분자로 표면 개질 되어서 활용되기도 하였음. 하지만 이러한 기존의 나노 섬유는 3차원 시트의 형태로 제작되었으며 3차원 조직을 재건하는데 한계점을 보임. 따라서 본 연구에서는 줄기 세포의 기능을 조절하기 위해 나노 섬유를 단편화 하여 제작된 세포 외 기질 모사체를 포함하는 스페로이드를 제작하였음. 먼저 poly (l-lactic acid) (PLLA) 시트의 아미노 분해를 통해 단편화된 섬유 (FF)를 얻었고, 세포 부착을 위해 폴리도파민 코팅을 하였음. 그 다음으로 섬유아 세포를 이용해 스페로이드 복합체를 제작하여 DNA 함량의 증가 및 높은 생존능을 확인하였음. 이와 마찬가지로 인간 비갑개 유래 줄기 세포 (human turbinate mesenchymal stem cell-
