Abstract
히아루론산 등 폴리 사카라이드 기반 하이드로젤의 바이오 응용은 매우 다양한 분야에서 이루어져 왔고, 앞으로도 지속적으로 연구될 분야이다. 그 이유는, 폴리 사카라이드의 우수한 생체 적합성을 기반으로, 화학적으로 다양한 분자와의 결합이 가능하고, 이에 따라 목적에 적합한 분자 설계가 가능하기 때문이다. 본 논문에서는, 이러한 장점들을 바탕으로, 폴라 사카라이드와 다양한 생체분자와의 결합을 통해, 여러 가지 목적에 부합하는 하이드로젤 복합체를 설계/제조하였고, 세포와 인공피부, 인비보 실험을 통해 재생의학적 관점에서 실질적인 활용성을 평가하였다. 특히, 화학적 가변성 (chemical versatility)을 활용하여, 화장품 및 의약 분야에서 실질적으로 응용이 가능한 플랫폼을 연구하는데 중점을 두었다. 1장에서는 히아루론산, 알진산 등 대표적인 폴리사카라이드와 이를 기반으로한 하이드로젤 등 베지클에 대하여 서술하였고, 조직공학과 약물 전달 관점에서의 응용에 대한 최근 연구동향을 다루었다. 2장에서는 소수화된 히아루론산 기반 나노 입자의 물리 화학적 특성을 조절하여, 경피 흡수가 가능한 수준으로 난용성 약물을 가용화하고, 세포 수준과 인공 피부에서, 담지된 약물이 효율적으로 전달되어 효능이 발현되는 신규 시스템을 제안하였다. 3장에서는, 폴리 사카라이드와 세포 부착성이 높은 RGD 펩타이드를 결합시켜 젤을 제조하고 줄기 세포 전달체로서의 특성을 확인하였으며, 인 비보 수준에서 조직이 생성됨을 확인하였다. 이를 통해 새롭게 설계된 하이드로젤의 재생의학적 가능성을 제시하였다. 4장에서는, 클릭 케미스트리를 통해 자기 조립이 가능한 새로운 형태의 폴리사카라이드 하이드로젤을 설계하여, 생체 주입이 가능한 세포 전달체가 제조 가능함을 인비트로와 인비보에서 조직 재생 관점에서 평가 하였다. 5장에서는, 약물의 전달을 극대화하기 위해, 하이드로젤과 바이러스 전달체의 복합체를 설계하였으며, 피부 암세포 치료를 위한 AAV mediated drug delivery 시스템에 대한 기초 연구를 진행하였다. 모델 약물인 6-gingerol이 AAV mediated drug carrier에 의해 암세포에 보다 효과적으로 전달되어 빠르게 사멸됨을 확인하였으며, gel coated viral vector 형태로 하이드로젤 내부에 보다 쉽게 봉입이 가능함을 제시하였다.; Polysaccharides such as hyaluronic acid (HA) are widely used in the design of engineered hydrogels, due to its biofunctionality, as well as numerous sites for modification with reactive groups. These hydrogels are widely used in the field of biomedical industry including transdermal delivery, regenerative tissue engineering and biosensor. In this thesis, I studied useful ways to fabricate polysaccharides based engineered hydrogels which can be used for drug delivery and regenerative tissue engineering. To enhance the drug delivery efficiency, i) the hydrophobically modified hyaluronic acid was designed, ii) particle assembly based on the complex of natural polymer resin and the hydrophobically modified hyaluronic acid were fabricated to prevent recrystallization of incorporated drugs, and finally iii) new drug solubilizing system for transdermal delivery was developed. In particular, the combination of hydrophobically modified hyaluronic acid and polymer resin contributed to suppressing crystallization of drug. Also, high drug delivery efficiency was obtained with the intrinsic property of hyaluronic acid. For the stem cells differentiation, bone morphogeneticprotein-2 (BMP-2) was loaded to Arg-Gly-Asp (RGD) peptide-conjugated alginate hydrogel. Also, the effects of BMP-2 delivery mode on the osteogenic differentiation of human bone marrow-derived mesenchymal stem cells (BM-MSCs) were investigated. Newly designed polysaccharide hydrogel can be used as a carrier of BMP-2 and to create a suitable environment for cell culture. Human BM-MSCs were induced by using a bottom-up and top-down release system and the cells were characterized in terms of alkaline phosphatase (ALP) activity and differentiation. The results may provide a useful tool for expanding the potential applications of stem cell therapy. Hyaluronic acid-based injectable hydrogel via copper-free click chemistry was developed and the clinical potential of newly designed hydrogel through in-vivo cartilage regeneration was evaluated. To that end, we first synthesized DBCO-modified HA via a simple EDC/NHS coupling reaction. The HA-PEG4-DBCO hydrogels were fabricated by mixing the 4-arm PEG azide. The clinical potential, cytotoxicity, biocompatibility and biodegradation rate were confirmed in in-vivo and in-vitro. In addition, cartilage regeneration using our hydrogel revealed the successful development of cartilage lacunae. Through this approach, we demonstrated the feasibility of our hydrogel for use as prototype cell delivery vehicles, which will be further refined in the future for particular biomedical applications in tissue engineering and other industries. The proposed systems, will hopefully contribute to numerous fields of drug delivery and tissue regeneration applications.
