dc.description.abstract조개풀(ArthraxonhispidusThunb.Makino)은벼과식물로한국,일본,중국그리고러시아에널리분포하고있으며천연염료의원료로많이사용되고있다.또한조개풀추출물은항염활성,천식,그리고알러지에효과가있다는연구결과가보고되었으며,flavonoid및phenoliccompound들이분리및보고되어있다.최근조개풀에서분리된C-glucosideflavonoid화합물들이유방암전이를저해한다는문헌들이보고되어있으며이러한문헌을통해선행연구를진행한결과,조개풀50%주정추출물에서유방암전이를억제하는활성이최대화가되는것을확인하였다.따라서본연구에서는조개풀50%주정추출물을대상으로실리카젤,다이아이온(HP-20)등다양한레진을이용한컬럼크로마토그래피및HPLC분취를통하여화합물을순수분리정제하였다.분리된성분들에대해서분광분석데이터(1Dand2DNMR,HR-QTOF-MS)를이용하여화학적구조를규명하였으며,알려진화합물10종[Apigenin8-C-β-D-glucopyranoside(vitexin)(1),Luteolin8-C-β-D-fucopyranoside(5),Apigenin8-C-α-D-fucopyranoside(6),Apigenin8-C-β-D-fucopyranoside(8),Luteolin(10),Medioresinol(11),Apigenin(13),Tricin(14),Tricin-4'-O-(7'',8''erythro-β-guaiacylglyceryl)ether(15),Tricin-4'-O-(7'',8''threo-β-guaiacylglyceryl)ether(16)]과신규화합물6종[(Apigenin-8-C-β-D-fucopyranosyl-4'-O-β-D-glucopyranoside(2),Luteolin-8-C-α-D-fucopyranoside(3),Luteolin-8-C-α-aceticacid(4),7-Methoxy-apigenin-6-C-α-L-fucopyranoside(7),7-Methoxy-apigenin-6-C-β-L-fucopyranoside(9),Tricin4'-O-(2-glyceryl)ether(12)]의구조를동정하였다.추후조개풀에서분리된화합물들의활성메커니즘을알기위한연구가필요하며,조개풀및C-glucoside유도체를이용한유방암전이및항염증질병의예방을위한신약후보로이용될수있다.|ArthraxonhispidusThunb.Makino,belongstothefamilyGramineae(Poceae)family, whicharewidelydistributedinKorea,Japan,China,andRussia.Thisplantisaperennial herbandcalled‘Kobunagusa’inJapaneseasasourceofnaturaldyestufffortraditional yellowsilkcloth.A.hispidushasbeenusedasherbalmedicinetoreducecough,asthma, allergy,andinflammatorydiseases.FewreportsofconstituentswereisolatedfromA. hispidusinhibitbreastcancermetastasis.Asaresultofconductingpreviousstudybasedon thisliterature,itwasconfirmedthatsuppressionofbreastcancermetastasisinA.hispidus extractswasmaximizedin50%ofethanolextract. Inpresentstudy,thecompoundsfrom50%ofethanolextractinA.hispiduswere isolatedusingseveralchromatographictechniquesandmassanalysistoyieldsixnew flavonoidderivatives[(Apigenin-8-C-β-D-fucopyranosyl-4'-O-β-D-glucopyranoside(2), Luteolin-8-C-α-D-fucopyranoside(3),Luteolin-8-C-α-aceticacid(4),7-Methoxyapigenin-6-C-α-L-fucopyranoside(7),7-Methoxy-apigenin-6-C-β-L-fucopyranoside(9), Tricin4'-O-(2-glyceryl)ether(12)],togetherwith10knowncompounds[Apigenin8-C-βD-glucopyranoside(vitexin)(1),Luteolin8-C-β-D-fucopyranoside(5),Apigenin8-C-α-Dfucopyranoside(6),Apigenin8-C-α-D-fucopyranoside(8),Luteolin(10),Medioresinol (11),Apigenin(13),Tricin(14),Tricin-4'-O-(7'',8''erythro-β-guaiacylglyceryl)ether(15), Tricin-4'-O-(7'',8''threo-β-guaiacylglyceryl)ether(16)].Thechemicalstructuresof compounds1–16wereelucidatedbasedonspectroscopicandMSdata(1Dand2DNMR, HR-QTOF-MS).Furtherstudiesarenecessarytoevaluatethebiologicalactivitiesofnewly isolatedcompoundsfromA.hispidus.Also,thedevelopmentofnewdrugcandidates shouldbeinvestigatedforthepreventionofcancerandinflammatorydiseasesbyCglucosidederivativesobtainedfromA.hispidus.
