Abstract
서 론: 현재 간 이식과 간 절제술로 인한 간 허혈-재관류(HIR) 손상은 좋은 치료와 예방 조치가 없다. 본 연구는 metformin의 간에 HIR손상 및 심장 손상에 대한 보호 작용을 연구 하고자 한다. 방 법: Wistar 수컷 쥐는 무작위로 대조 팀, 허혈-재관류 팀(IR팀), metformin 150 mg / kg팀 과 metformin 100 mg / kg 팀으로 나뉜다.Wistar 수컷 쥐 수술 전 30min에게 metformin 100 mg / kg, 150 mg / kg, 식염수를 각각 건넨다.(n = 4) 결 과: metformin은 HIR가 일으키는 간의 손상과 심근 손상을 줄일 수 있다.그 결과에 따르면 IR팀 혈청alanine transaminase (ALT), 텐 동 aspartate transaminase (AST), malondialdehyde (MDA)는 metformin 치료 팀보다 훨씬 높은 것으로 나타났다. IR팀은 간 catalase (CAT)와 Superoxide dismutase (SOD)의 활성화는 metformin 치료 팀보다 현저히 낮다. IR팀 심장 malondialdehyde (MDA), creatine kinase MB (CK - MB), lactate dehydrogenase (LDH)의 활성화는 metformin 치료 팀보다 현저히 높고 catalase (CAT)의 활성화는 metformin 치료 팀보다 현저히 낮다. 결 론: metformin은 산화 스트레스를 억제하여 HIR가 일으키는 간의 손상과 심근 손상을 줄일 수있다.|Background: At present, there is no good treatment and prevention for hepatic ischemia-reperfusion (HIR) injury caused by liver transplantation and hepatectomy. The aim of this study was to investigate the protective effect of metformin on hepatic ischemia-reperfusion injury and related cardiac injury in rats. Methods: Wistar male rats were randomly divided into four groups (n= 4) such as sham group, IR group, IR treated with metformin 150mg/kg, and IR treated with metformin 100mg/kg. Wistar male rats were administrated with metformin in 100mg/kg, 150mg/kg, and saline before operation 30min and suffered from 15min ischemia and 6h reperfusion. (n=4) Results: We found that metformin significantly alleviate hepatic ischemia-reperfusion injury and myocardial injury caused by HIR. The results showed that the levels of alanine transaminase (ALT) and aspartate transaminase (AST) of serum and malondialdehyde (MDA) in IR group were significantly higher than those in metformin-treated group, enzymes activity of catalase (CAT) and superoxide dismutase (SOD) in IR group were significantly lower than that of metformin-treated group in the liver. The levels of malondialdehyde (MDA), creatine kinase MB (CK-MB), and lactate dehydrogenase (LDH) activity in IR group were significantly higher than those in metformin-treated group, and catalase (CAT) activity was lower than that in treatment group in the heart. Conclusion: Metformin can alleviate hepatic ischemia-reperfusion injury and myocardial injury induced by HIR through inhibiting oxidative stress reaction.
