Abstract
항산화 효소는 생물체 내에서 대사 과정을 통해 생성되는 ‘고반응 산소종 (reactive oxygen species, ROS)’의 산화적 스트레스에 대한 중요한 방어 기작 중의 하나이다. 생체 내에서 활성 산소를 생성하는 환경 오염 물질의 산화적 스트레스와 관련하여, 해양의 조간대에 주로 서식하는 Tigriopus japonicus 에서 항산화 작용에 관여하는 유전자들의 구조와 발현 양상을 조사하였다. 이를 위하여, T. japonicus로부터 superoxide dismutase(Cu/Zn, MnSOD), Catalase (CAT), glutathione S-transferase omega (GSTω) 유전자의 전체 염기서열을 분석 하고, 이들의 아미노산 서열은 기존에 보고된 다른 척추동물과 무척동물과의 유사도 분석을 위해 이용되었다. 이들 유전자의 open reading frame (ORF) 와 아미노산의 길이는 각각 Cu/Zn SOD 705bp (235 aa), MnSOD 681bp (227 aa), Catalase 1590bp (530 aa), GST ω 771bp (257 aa) 였다. 각각의 유전자로부터 암호화되는 단백질의 분자량과 pI 값은 Cu/ZnSOD 25.3kDa (6.41), MnSOD24.5 kDa (6.75), catalase 58.3 kDa (7.31), GSTω 29,7 kDa (5.66)으로 측정되었다. 또한 T. japonicus로부터 얻은 각각의 유전자들은 척추, 무척추동물의 유전자들과의 유사성을 조사하기 위하여 ClustalX 1.83를 이용하여 alignment하였다. T. japonicus의 4개의 항산화 유전자 중에서 MnSOD아미노산 염기서열은 Homo sapiens (61%), Danio rerio (64%), Bombyx mori (63%), Caenorhabditis elegans (65%) 순으로 높은 유사성을 보였으며, GSTω 아미노산 염기서열은 기존에 보고된 Homo sapiens(30%), Xenopus tropicalis (32%), Takifugu rubripes (31%), Halocynthia roretzi (26%), Crassostrea gigas (32%), Bombyx mori( 27% )의 GST 서열과 상당히 낮은 유사성을 보여 주었다. 다양한 중금속 (Zn, Cu, Hg, Ag, Mn, Ni, )과 과산화수소 (H₂O₂)에 의한 산화적 스트레스에 따른 이들 항산화 유전자의 발현 변화는 real time RT-PCR 이용하여 측정하였다. Tigriopus Cu/Zn SOD 유전자의 발현은 모든 중금속 노출의 초기에 저해되는 양상을 보였고, 시간이 지남에 따라 발현 양이 점차 회복되는 경향을 보였다. 또한 Tigriopus catalase 유전자 발현은, Mn (50 and 100 μg /L)를 처리한 경우, 농도 의존적인 감소 경향을 보인 반면, 과산화수소 (H₂O₂, 2mM)를 시간 별로 처리하였을 때는 발현 양이 지속적으로 증가되는 양상을 보였다. 따라서, 본 연구 결과는 수계에 유입된 다양한 중금속이 T. japonicus의 항산화 관련 유전자들의 발현에 영향을 미칠 수 있음을 의미한다.; Contaminant-related changes in antioxidative process in the intertidal harpacticoid copepod Tigriopus japonicus exposed to model redox cycling contaminants were assessed. A number of antioxidant genes involved in detoxification was cloned using degenerated primers. We obtained full-length cDNA of four antioxidant genes; superoxide dismutase (Cu/Zn SOD, MnSOD), catalase (CAT) and glutathione S-transferase omega (GST ω) by 5' and 3' rapid amplification of cDNA. The resulting sequences contained open reading frames (ORF) of 705bp (235 aa) for Cu/Zn SOD, 681bp (227 aa) for MnSOD, 1590bp (530 aa) for CAT, and 771 bp (257 aa ) for GSTω . The four antioxidant cDNAs encode protein with 25.3kDa (Cu/ZnSOD), 24.5kDa (MnSOD), 58.3 kDa (CAT), and 29.7kDa (GSTω), respectively. The deduced amino acid sequence showed theoretical pI of 6.41 for Cu/Zn SOD, 6.75 for MnSOD, 7.31 for CAT, and 5.66 for GSTω, respectively. After motifs search, we found that Cu/Zn SOD signature in GNAGsRlACgvI (217 - 228), Mn and Fe SOD signature in DvWEHAYY (189-197), and catalase proximal active site signature in FdReripERvvHakGAG (64-81), respectively. We measured mRNA expression pattern of these genes from T. japonicus exposed to different heavy metals (Zn, Cu, Hg, Ag, Mn, and Ni) and hydrogen peroxide (H₂O₂). The expression pattern of Cu/Zn SOD gene induced by each metal showed down-regulation at first and then getting back to adjust in changed environmental condition. Catalase expression showed a concentration-dependent decrease when T. japonicus was exposed to Mn (50 or 100ug/L). However, in case of H2O2 exposure (2 mM), a time-dependent increase was observed in catalase mRNA expression. Our results provide some basis for understanding the antioxidant gene defense mechanisms in an ecologically- and phylogenetically important species (T. japonicus). These results will also enable us to make a comparison of defense mechanisms of T. japonicus with other marine organisms.
