Abstract
유전자 전달체인 High mobility group box 1 (HMGB1) 은 핵 단백질로서, 이 중가닥의 DNA와 결합하는 특성을 가지고 있다. HMGB1은 box A, box B, 산성 아 미노산으로 이루어진 도메인 등 총 3개의 도메인으로 구성되어 있다. 본 연구에 서는 유전자 재조합 기술을 이용하여 재조합 HMGB1 펩타이드 전달체를 개발하고, 그 특성에 관하여 연구하였다. 먼저 human HMGB1 cDNA 얻기 위하여 293세포 에 서 추출한 RNA를 주형으로 하여 RT-PCR을 수행하였다. 증폭한 HMGB1를 대장균에 서 발현시키기 위하여 pET21a-rHMGB1의 재조합 플라스미드를 구축하였다. HMGB1 의 C-말단에는 산성 아미노산 부분이 다량 존재하며, 이 부분의 음전하가 HMGB1 과 DNA의 결합을 저해하므로 산성 아미노산 부분을 제거하여 보다 효율적인 펩 타이드 전달체를 개발하였다. 아미노산 말단에 히스티딘이 연결된 pET21a 벡터 에 증폭한 HMGB1 유전자를 삽입한 재조합 플라스미드 (pET21a-rHMGB1)를 대장균 BL21에 형질전환 시켜 IPTG로 유도한 후, rHMGB1를 니켈 친화성 크로마토그래피 로 순수하게 분리·정제하였다. 순수하게 정제된 rHMGB1와 DNA간의 복합체 형성 을 gel retardation assay를 통하여 확인한 결과 1:1 (pDNA:rHMGB1) 무게 비율 에서 완전히 지연되었으며, 동적 광 산란 분석법을 통하여 이 복합체의 크기가 193nm임이 측정되었다. 293 세포에서 rHMGB1와 rHMGB1/pDNA 복합체의 독성을 조 사한 결과 독성이 전혀 나타나지 않았고, 대조구인 poly-L-lysine (PLL,20kDa) 은 매우 높게 나타났다. rHMGB1의 유전자 전달 효율 조사는 1:40에서 가장 최적 비율로 나타났고, 이 비율에서 대조구인 PLL보다 높은 transfection 효율을 보 였다. 최근 rHMGB1의 box B 부분이 초기 염증반응을 일으킨다는 보고가 있다. 본 연구에서는 보다 안전하고 효율적인 유전자 전달을 위하여 염증반응을 일으 키는 box B와 DNA와 결합력을 저해하는 C-말단의 산성 아미노산 부분을 제거하 였다. 그리고 높은 유전자 전달을 위하여 N-말단에 HIV-1유래 TAT 유전자를 융 합하여, 재조합 TAT-HMGB1 box A (rTAT-HMGB1A) 펩타이드 유전자 전달체를 생산 및 정제, 그 특성에 관한 연구를 하였다. 발현 벡터인 pET21a-rTAT-HMGB1A를 구 축한 후, IPTG에 의해 rTAT-HMGB1A를 발현시키고 니켈 친화성 크로마토그래피를 통하여 정제하였다. 또한 TAT가 제거된 재조합 HMGB1A (rHMGB1A)를 발현 및 정 제를 하여 대조구로 이용하였다. 펩타이드와 DNA간의 복합체 형성을 gel retardation assay를 통하여 확인한 결과 모두 1:2에서 완전히 지연되었다. rTAT-HMGB1A은 rHMGB1A 에 비하여 높은 유전자 전달 효율을 보여줄 뿐만 아니라, 293세포에서 독성이 전혀 나타나지 않았다. 이와 같은 결과들로 미루어 볼 때, rHMGB1와 rTAT-HMGB1A 는 적절한 유전자 전달 효율을 가지며, 독성이 전혀 없는 장점이 있어 세포 내 유전자 전달체로 유용하게 이용되어질 수 있다고 사료되어 진다.; High mobility group box 1 (HMGB1) is an abundant nuclear protein, which binds to double stranded DNA. HMGB1 is composed of high mobility (HMG) box A, box B, and C-terminal acidic region. In this study, a recombinant high mobility group box 1 (rHMGB1) protein was produced and characterized as a carrier of nucleic acids. First, the human HMGB1 cDNA was cloned by RT-PCR. The rHMGB1 expression vector, pET21a-rHMGB1, was constructed with the cDNA. In pET21a-rHMGB1, the acidic box of the c-terminus of the wild type HMGB1 was eliminated, since it had highly negative charges and may have interfered with the DNA and peptide interaction. pET21a-rHMGB1 was transformed into the BL21 strain and rHMGB1 was produced by IPTG induction. The resulting rHMGB1 was purified using a nickel column. A gel retardation assay showed that plasmid DNA (pDNA) was completely retarded at a 1:1 weight ratio (pDNA:rHMGB1). The complex size measured by dynamic light scattering was approximately 193 nm. The cytotoxicity of rHMGB1 was compared with poly-L-lysine (PLL, 20 kDa) as a peptide or peptide/pDNA complex. As a result, rHMGB1 did not show any cytotoxicity to 293 cells, while PLL had a much greater cytotoxicity. rHMGB1 had the highest transfection efficiency at a 1:40 weight ratio (pDNA:rHMGB1). In addition, rHMGB1 showed higher transfection efficiency than PLL. Recent reports showed that HMGB1 is a pro-inflammatory cytokine, with the pro-inflammatory domain located in box B. Here, an HMGB1 peptide was genetically engineered to produce a recombinant HMGB1 (rHMGB1) peptide containing only the Box A region (rHMGB1A). Box B and the acidic domain were removed for safe gene delivery without the pro-inflammatory effects but with efficient complex formation, respectively. Additionally, a TAT peptide-rHMGB1A fusion peptide (rTAT-HMGB1A) was created for more efficient cell entry. It was located at N-terminus of HMBG1 box A (HMGB1A). A recombinant TAT-HMGB1A peptide was expressed, purified, and characterized as a carrier of nucleic acids. The HMGB1A cDNA was amplified by PCR. The expression vector, pET21a-rTAT-HMGB1A, was constructed with the cDNA. rTAT-HMGB1A was overexpressed by the IPTG induction and purified using nickel affinity chromatography. A recombinant HMGB1A (rHMGB1A) without TAT domain was also overexpressed and purified as a control. In a gel retardation assay, both HMGB1A peptides formed complexes with DNA equally well. The transfection assay showed that rTAT-HMGB1A had higher gene transfer efficiency than rHMGB1A. In addition, rTAT-HMGB1A had no cytotoxicity to 293 cells. In conclusion, with low cytotoxicity and moderate transfection efficiency, rHMGB1 and rTAT-HMGB1A have a potential to be used as an efficient non-viral carrier for the nucleic acid delivery.
