dc.description.abstract많은 환자들이 예기치 않은 사고나 질병으로 인한 조직 또는 장기 손상으로 많은 고통을 겪고 있다. 이를 해소하기 위해 장기 이식이 이루어 지고 있으나 기 증자 수의 제한으로 모든 환자를 치료하기에 충분하게 이루어지지 않고 있는 실 정이다. 조직공학은 장기 이식을 대체할 수 있는 대체 치료법으로 각광받고 있다. 주사 가능한 하이드로젤은 조직공학 분야에서 매력적인 재료이다. 주사 가능한 방법은 최소한의 외상의 치료로 전달하거나 이식하는 것이 가능하다. 이 연구에 서, 우리는 화학적 가교제를 사용하지 않고 세포를 이용하여 가교한 알긴산 하이 드로젤을 만들었다. 세포부착성 펩티드가 결합된 알긴산은 세포와의 특이적 상호 작용을 통해 가교된다. 우리는 유변학적 측정을 통해 세포로 가교되어 젤이 되는 거동을 살펴보았다. 기계적 물성은 세포와 칼슘 이온의 양으로 조절될 수 있음을 확인하였다. 또한, 동물 실험을 통해 세포로 가교된 하이드로젤의 연골 재생 효 과에 대해 살펴보았다. 재생된 조직은 H&E, Safranin O 염색을 통해 분석하였으며, FRET과 FCS를 통해 세포와 고분자 혼합물에서 정량적 정보를 얻을 수 있었다. 세포를 이용한 가교 방법은 조직공학적으로 유용한 지지체를 만들 수 있으며, FRET과 FCS 방법을 이용하여 고분자와 세포간 특이적 상호작용을 측정할 수 있 다.; Many patients suffer from a loss or failure of tissue or organ caused by unexpected accidents or disease. Organ transplantation is one typically adapted to treat these patients. However, the number of donors has been extremely limited. Tissue engineering is one recent approach to provide man-made tissue or organ. Injectable hydrogels are attractive carriers for tissue engineering, as gels can be used for delivery of cells with minimal invasive surgical treatment. In this study, we prepared injectable hydrogels using cell-interactive polymers via cell cross-linking without chemical cross-linking reagents. RGD-modified alginate solution was cross-linked with cells by specific interactions between receptors in the cells and ligands coupled to alginate. We confirmed gelation behavior of cell cross-linked hydrogels by rheological measurement. The more Ca2+ and cells added to cell-polymer mixture, the shorter the gelation time and the higher the mechanical properties of cell cross-linked hydrogels were obtained. We also found that the mechanical properties of cell cross-linked gels could be controlled by the concentration of cells and calcium ions. These gels were useful to deliver primary chondrocytes to mice in a minimally invasive manner, resulting in the remarkable formation of cartilage tissues in vivo. We also quantified interactions between polymer and cells by FRET and FCS. This approach to cross-linking polymers with cells could be useful for tissue engineering. In addition, FRET and FCS provided a useful means to investigate specific cell-polymer interactions in vitro.-
