Abstract
Augustine에 의해 제안된 담지법(이하 Augustine 담지법)을 적용하여 키랄 Ru-BINAP 촉매를 anchoring agent인 phosphotungstic acid (PTA)로 개질된 알루미나 담체에 담지 하였다. 담지 촉매의 특성분석은 FT-IR, ^(31)P MAS NMR 및 ICP-AES를 사용하여 분석하였는데 PTA와 Ru-BINAP이 개질담체 위에서 안정적으로 결합을 형성함을 알 수 있었다. 담지 촉매들의 촉매 성능은 dimethyl itaconate (DMIT)의 비대칭 수소화 반응을 통해 조사하였다. 담지 촉매의 활성과 광학선택도는 균일계 촉매에 비해 약간 떨어졌지만 기질/촉매(S/C) 비율을 크게 증가시켜도 안정적인 반응속도를 나타내었다. 촉매 리간드의 형태와 촉매 담지 온도는 촉매 성능에 크게 영향을 미침을 알 수 있었다. 또한 반응 온도, 수소 압력, 용매 그리고 첨가제의 종류 및 양의 최적화에 의해 활성과 광학선택도를 크게 향상 시킬 수 있음을 알 수 있었다. Augustine 담지법을 변형하여 PTA/Al₂O₃ 담체를 제조하였고, 이 변형된 담체에 Ru-BINAP 담지 촉매를 제조하였다. 이들 제조된 촉매를 methyl acetoacetate (MAA) 비대칭 수소화 반응에 적용하였고, 또한 Augustine 담지법으로 제조된 촉매의 결과와 비교하였다. 변형된 Augustine 담지 촉매의 특성은 FT-IR, XRD 및 ICP-AES를 이용하여 분석하였다. 변형된 Augustine 담지 촉매는 Augustine 촉매에 비해 MAA 비대칭 수소화 반응에서 휠씬 빠른 반응 전환율과 더 좋은 선택도 및 광학선택도를 나타내었다. 이러한 특징들은 촉매 제조에서 사용되는 용매들의 차이에 의한 촉매들의 서로 다른 표면 특성에 의해 설명 할 수 있다. 변형된 Augustine 담지 촉매를 다른 종류의 β-ketoester 유도체들의 비대칭 수소화 반응에도 적용하였는데, 균일계 Ru-BINAP 촉매계의 결과와 같이 높은 수율과 광학선택도를 갖는 β-hydroxyester를 제조할 수 있었다. 변형된 Augustine 담지 촉매의 상업적 응용을 위한 높은 TON에서의 반응에서도 유사한 촉매 활성과 우수한 광학선택도를 나타내었다. 변형된 Augustine 담지 촉매의 재사용 가능성을 확인하기 위해 MAA 수소화 반응을 5회 반복 수행 하였는데 사용 횟수가 증가함에 따라 반응 전환율과 선택도가 점진적으로 감소 되었지만 광학선택도는 거의 유사한 값을 보였다. 변형된 Augustine 담지촉매는 MAA 비대칭 수소화 반응에서 공정변수에 따라 경향을 달리 하였는데, 반응 온도가 증가함에 따라 상당한 반응속도와 선택도 및 광학선택도의 변화를 보였지만, 반응 압력은 크게 영향을 미치지 않았다. Augustine 담지법을 동적 반응속도론적 분할(DKR) 반응에 확대 적용하여 보았다. 라세미화 촉매인 Ru-p-cymene을 Augustine 담지법으로 담지 하였고, 이 촉매와 효소를 혼합하여 1-phenylethanol의 DKR 반응을 실시하고 그 결과들을 균일계 촉매의 결과들과 비교하였다. 담지 촉매는 균일계 촉매에 비해 높은 수율과 우수한 선택도 그리고 높은 촉매 활성을 나타내었다. 담지 촉매의 재사용 실험에서는 전환율은 약간 감소하였지만 선택도의 변화 없이 여러회 재사용 할 수 있었다. 또한 DKR 반응의 주요 공정 변수에 따른 담지 촉매의 특성을 조사하였다. 공정변수의 대한 영향은, 온도가 높아지고, 촉매/효소 비율이 커질수록 담지 촉매가 라세미화를 촉진 하였으며 높은 TON에서도 안정적인 촉매 활성을 나타내었다. 또한 담지 촉매는 공정 변수의 변화에 관계없이 높은 광학순도의 생성물 에스터를 얻을 수 있었다.; Chiral Ru-BINAP complexes were immobilized on alumina support using a phosphotungstic acid (PTA) as the anchoring agent, via a method suggested by Augustine (Augustine method). The immobilized catalyst was characterized by FT-IR, ^(31)P MAS NMR and ICP-AES, and it was found that a stable bond was formed between PTA and Ru-BINAP on the modified support. The catalytic performance of the immobilized catalysts was investigated in asymmetric hydrogenation of dimethyl itaconate (DMIT). The activity and enantioselectivity of the catalysts were slightly lower than those of the homogeneous ones but the immobilized catalyst showed a stable reaction rate even when the substrate/catalyst (S/C) ratios was increased greatly. The type of catalyst ligand and the temperature for catalyst immobilization were found to affect the catalyst performance. It was also found that the activity and enantioselectivity could be significantly enhanced by optimizing the reaction temperature, hydrogen pressure, solvents and additives. PTA/Al₂O₃ support was prepared by modification of Augustine method, and Ru-BINAP on this modified support was immobilized (Modified Augustine method). The prepared catalysts were applied in the asymmetric hydrogenation of methyl acetoacetate (MAA) and their catalytic performance were compared with those of catalyst prepared by Augustine method. The modified Augustine catalyst was characterized by FT-IR, XRD and ICP-AES. The modified Augustine catalyst showed faster conversion and better selectivity and enantioselectivity than those of Augustine catalyst. These results could be explained by the different surface charateristics of catalysts obtained by using the different solvents in catalyst preparation. The modified Augustine catalyst was applied in asymmetric hydrogenation of various β-ketoester derivatives, and they could produce β-hydroxyesters with good yield and enantioselectivity, similar to the results obtained in homogeneous Ru-BINAP catalyst systems. In the high TONs test for modified Augustine catalyst, the similar activity and good enantioselectivity were obtained. The possibility of recycling for modified Augustine catalyst was tested. The gradual decrease in conversion and yield of product were observed after several subsequent runs, but enantioselectivity was constant in each subsequent runs. The effect of reaction variables on modified Augustine catalyst were changed. Although the reaction rate, selectivity and enantioselectivity of product were changed depending on the reaction temperature, the changes of hydrogen pressure did not affect the catalytic performance. An Augustine method has extended to the dynamic kinetic resolution (DKR). Ru-p-cymene as a catalyst for racemization was immobilized on alumina support by Augustine method, and this catalyst with enzyme were used in the DKR of 1-phenylethanol and its performance was compared with that of homogeneous ones. The immobilized catalyst showed higher activity, better yield and selectivity than those of homogeneous ones. The immobilized catalyst could be reused in several subsequent runs without any significant loss of selectivity to product. The effect of reaction variables on the performance of immobilized catalyst were also investigated. As reaction temperature and catalyst/enzyme ratios were increased, it accelerated racemization rate by immobilized catalyst. Lastly, the immobilized catalyst could produce esters with high optical purity independent of reaction variables.
