Abstract
Background and Aims: It is well known that one of the 5-HT4 agonists, mosapride citrate affects gastric motiltiy. However, whether mosapride citrate have any effects on the visceral pain of colon or rectum is not sure. The aims of this study were to assess the effects of mosapride citrate on the visceral pain in a rat visceral hypersensitivity model. Methods: Perception of visceral pain was evaluated by the visceromotor response (VMR) to colorectal distension (CRD) observed on the electromyographs (EMG) from the abdominal musculature in the urethane anesthetized rats. Visceral hypersensitivity was induced by intrarectal instillation of 4% acetic acid or 1.5 % zymosan. Mosapride citrate was administerated intraperitoneally 3 hours later. VMR to CRD were recorded prior to acetic acid or zymosan instillation and before and after mosapride citrate treatment. Results: Intraperitoneal administration of mosapride citrate increased gastric contraction at the dose of 5mg/kg. Intracolonic instillation of acetic acid significantly increased EMG responses of the abdominal muscles to CRD compared to pretreatment state. (100 %) (174±24 %, n=8, P < 0.05) Intracolonic instillation of zymosan significantly increased EMG responses of the abdominal muscles to CRD compared to pretreatment state. (100 %) (144±9 %, n=8, P < 0.05) Mosapride citrate, intraperitoneally injected, significantly reduced EMG responses (100%) increased by acetic acid instillation at the dose of 5mg/kg. (61±9 %, n=8, P<0.05) Mosapride citrate, intraperitoneally injected, significantly reduced EMG responses (100%) increased by zymosan instillation at the dose of 5mg/kg. (67±9 %, n=8, P<0.05) Conclusions: 5-HT4 receptor agonist, mosapride citrate, significantly reduced VMR to CRD in acetic acid- and zymosan-induced IBS model at the dose of 5mg/kg. Therefore, mosapride citrate might have effects on the pain of visceral hypersensitivity model induced with acetic acid or zymosan.
