Abstract Functional studies on the cnp-2 in Caenorhabditis elegans Jung, Hana Department of Life Science The Graduate School Hanyang University Calcineurin is a serine/threonine phosphatase implicated in a wide variety of biological responses. Calcineurin consists of two subunits, catalytic subunit A (TAX-6) and regulatory subunit B (CNB-1). Previously, calcineurin interacting proteins have been screened by yeast two-hybrid assays. In the screening, we found that CNP-2 interacted with TAX-6 in vivo, and we showed that CNP-2 binds to the v catalytic domain of TAX-6. Phenotypes of calcineurin mutants are short body length, egg laying defect and reduced brood size. Therefore, we investigated function of cnp-2 in the calcineurin-mediated signal pathway. We have generated several double mutants between cnp-2 mutants and calcineurin mutants. Then, we characterized phenotypes associated with calcineurin mutants. The cnp-2 gene is expressed in head neuron, posterior intestine, spermatheca, distal spermatheca and spermathecauterine valve. Phenotypes of cnp-2 mutant alone seemed to be normal. Phenotypes of cnp-2 mutant in calcineurin mutant background indicated that calcineurin and cnp-2 might be related to brood size, egg laying, body length and defecation behavior in parallel pathway. In pharyngeal pumping behavior, we suggest that cnp- 2 functions as a modulator of calcineurin in the control of pharyngeal pumping.
