dc.description.abstractNurr1은 orphan nuclear receptor로 도파민 신경세포의 발달에 중요한 역할을 하고 성체의 중뇌에서 발현된다. 또한 Nurr1은 Neural precursor cell (NPC)s에서 도파민 신경세포의 생존을 증진하며, 유해한 자극에 견딜 수 있게 한다. Nurr1으로 형질 전환한 NPCs를 줄무늬체에 이식을 하게 되면 이식이 된 NPCs는 도파민 신경세포를 생성하고 이식된 뇌에 정착하여 이식조직을 생성하게 된다. 그러나 Nurr1으로 형질 전환한 NPCs에서 생성된 도파민 신경세포는 일시적으로만 발현이 되고 흰쥐에 이식을 실시할 경우 오랜 시간이 지나게 되면 도파민 신경세포는 지속되지 않고 감소하게 된다. 이에 우리는 이식을 할 NPCs에 constitutive active (ca)-Raf를 Nurr1과 동시발현하여 Raf-ERK signaling을 활성화시켜 Nurr1의 발현이 오랫동안 유지되도록 하여 다량의 도파민 신경세포를 가진 세포괴(cell graft)을 생성하였다. 이는 ca-Raf와 Nurr1을 같이 발현하게 되면 mRNA 전사의 지속적인 유지, 세포증식 및 세포증식 증진 등에 의해 Nurr1발현이 안정화되어 도파민 신경세포의 생존과 증식을 증진 시키는 것으로 확인하였다.|Nurr1 is an orphan nuclear receptor expressed in dopamine (DA) neurons of developing and adult midbrains. Engineering of Nurr1 in NPCs generates DA neurons with promoted cell survival and resistant to toxic stimuli. Intrastriatal transplantation of Nurr1-transduced NPCs generates DA cells survived and integrated into host brain. However, the enrichment of DA cells in the grafts generated by Nurr1-transduced NPCs was only transient but not sustained for long period of post-transplantation. We show herein that activation of Raf-ERK signaling in these donor cells by co-expression of constitutive active (ca)-raf contribute to a faithful generation of large grafts fully enriched with DA cells. The ca-raf co-expression effect was achieved by multiple mechanisms such as maintenance of Nurr1 protein stability, Nurr1 mRNA expression, and cell survival and proliferation.; Nurr1 is an orphan nuclear receptor expressed in dopamine (DA) neurons of developing and adult midbrains.
