Abstract
Much attention has been focused recently on electrospinning/-spraying methods for nano-/microparticle preparation due to their better characteristics than conventional methods. Ad-vantages of electrospinning/-spraying methods for the development of therapeutic particular sys-tems involve high yield of products, simple procedure and availability of scale-up. In order to de-velop optimized electrospun particles containing a water-insoluble anti-cancer drug, 17-AAG, var-ious experimental parameters were tested, such as single-gauge jetting vs. co-jetting, choice of biodegradable polymer, and electrospinning parameters. Core-shell type electrospun PCL nano/micro-particles and single electrospun PLGA nano/micro-particles containing 17-AAG were prepared through optimization process. Homogeneous size distribution and spherical shapes were obtained. Controlled in vitro drug release rates could be achieved by selecting proper polymer and modifying electrospinning parameters. Also, the anti-cancer effect was confirmed with the in vitro cytotoxicity measurement. Taken these data together, electrospun biodegradable nano/micro-particles provide a potential as carrier systems for efficient anti-cancer drug delivery in vivo.
