dc.description.abstractAcute myocardial infarction (AMI) is becoming one of the reasons of unexpected sudden deaths in the world. Since AMI brings fatal effects in the body, early detection is crucial. For the early diagnosis of AMI, a number of biomarkers have been developed. Among them, troponin I (cTn I) is a promising candidate of myocardial injury due to its great specificity and sensitivity regarding acute myocardial cell damage. This study reports detection of the cTn I biomarker using a surface-enhanced Raman scattering (SERS)-based immunoassay technique. Gold nanoparticles (Au NPs) and magnetic beads were used as detection probes and as capturing substrates, respectively. For this purpose, cTn I monoclonal antibodies (mAbs) were immobilized on the magnetic bead substrates, and then cTn I antigen solution and cTn I mAb-conjugated Au NPs were sequentially added to make the sandwich immunocomplexes. Because its entire reactions occur on the surface of the three dimensional substrates in solution, the SERS-based immunoassay technique clears the limitation in diffusion kinetic on a polar substrate. As a result, we can reduce assay time and obtain more reproducible SERS signal by measuring for the average nanoparticle ensembles in solution. Also, we can detect quantitative analysis of cTn I biomarker by monitoring variation of the SERS intensity in accordance with the number of sandwich immunocomplexes. The proposed method can be a potential clinical tool for the fast diagnosis of AMI by detecting cTn I biomarker.-
