Abstract
Extracellular matrix (ECM) is used as a biological scaffold material for regeneration of tissue, because scaffolds composed of ECM which consists of various proteins and bioactive molecules enhance regeneration of tissue. In this study, we designed ECM based thermosensitive hydrogel as a biological scaffold for medical applications. The soluble extracellular matrix (sECM) was successfully extracted from human adipose tissue through two major steps: i) solid ECM extraction from human adipose tissue and ii) sECM isolation from solid ECM by a protein denaturation method. The ECM-MC solutions were prepared by blending sECM and methylcellulose (MC) solutions at low temperature and the ECM-MC solutions showed a reversible sol-gel phase transition as temperature changes. The hASC-incorporated ECM-MC hydrogels accelerated wound healing in vivo. These results suggest ECM-MC hydrogels can serve as biological scaffolds for tissue regeneration and injectable carriers to deliver drugs or cells.
