- 본 연구에서는 carbocyclic nucleoside 유도체를 이용하여 방사성동위원소를 표
지하여 분자영상용 방사성의약품 개발에 적용하고자 하였다. 일반적인
nucleoside 유도체들은 sugar ring의 oxygen에 의해 체내에서 가수분해가 잘 일
어나 안정성이 떨어지는 단점이 있다. Carbocyclic nucleoside의 도입으로 in vivo
에서 구조적 안정을 추구하였으며, radioiodine labeled cis-1-[4-(hydroxymethyl)-2-
cyclopenten-1-yl]-5-[124I]iodouracil (carobcyclic [124I]d4IU)를 이용하여 MCA
hepatoma cells과 wild type MCA cells에서 HSV-1 TK (herpes simplex virus type-1
thymidine kinase) 유전자의 발현 영상을 측정하였다. 합성은 cyclopentadiene을
출발물질로 하여 Pd(0)-catalyzed coupling reaction 및 radioiodination을 주요 반응
으로 이루어졌다. Tributylstannyl group이 치환된 전구물질과 Na124I를 이용하여
radioiodination 반응을 수행하였고, radio-TLC를 통해 반응을 monitoring하였으며,
얻어진 product는 reverse phase HPLC system에 의해 분리 및 정제하였다.
Radioiodinated carobcyclic [124I]d4IU의 세포 내 섭취율에 대한 실험에서는 MCATK가
2배 이상 높은 것으로 확인되었다. Carbocyclic d4IU와 5-iododeoxyuridine
에 대한 생체 내 안정성을 테스트 하기 위하여 mouse liver microsome과 mouse
serum에서 테스트하였다. Carbocyclic d4IU는 5-iododeoxyuridine에 비해 mouse
liver microsome에서 3배 이상, mouse serum에서는 7배 이상 안정하다는 결과를
얻었다. MicroPET를 이용한 동물 영상 실험에서는 HSV-1 TK가 발현된 MCATK
종양을 선택적으로 영상화할 수 있었다. 이와 같은 결과를 토대로 cytosine
계열의 carobcyclic [124I]d4IC와 carobcyclic [124I]d4IVC를 합성하였다. carobcyclic
[124I]d4IC의 합성은 tributylstannyl group이 치환된 전구물질을 이용하였으며,
carobcyclic [124I]d4IVC의 합성을 위해서는 trimethylsilyl group을 도입하였다.
Radioiodination은 carobcyclic [124I]d4IU와 동일한 방법으로 Na124I를 이용하여
radioiodination 반응을 수행하였다. [18F]fluoroalkyl nucleosides의 carbocyclic 유도
체를 합성하기 위하여 기준물질인 carbocyclic fluoroalkyl nucleosides의 합성을
수행하였다.
