Especially, the network hub has been considered one of the most important topological properties to interpret a network as a complex system. However, previous structural brain network studies have reported network hub regions based on various nodal resolutions and an assumption that information flow of network is only spread through shortest connection. The aim of the dissertation is proposing the novel procedures to supplement problems of structural brain hub analysis. First, I hypothesized that brain network hubs should be determined considering various nodal scales in a certain range. I tested the hypothesis using the hub strength determined by the mean of the “hubness” values over a range of nodal scales. Some regions of the precuneus, superior occipital gyrus, and superior parietal gyrus in a bilaterally symmetric fashion had a relatively higher level of hub strength than other regions. These regions also had a tendency of increasing contributions to local efficiency than other regions. This framework provides a benefit in the detection of important brain regions in the network without any specific network nodal resolution. Second, to avoid the assumption that information flow of network is only spread through shortest connection, I provided the methodological pipeline for finding influential nodes of human brain network based on the Physarum centrality. It was possible to find optimal paths connecting two food sources by controlling amount of fluid flow and competing paths in the tubular network without the shortest path assumption. To the best of my knowledge, this is the first study employing Physarum model to investigate network hubs in the structural brain network. This pipeline consisted of two steps: 1) construction of brain network using diffusion tensor imaging, 2) apply Physarum model to brain network. Note that most of the network hub regions based on various centrality measures almost overlapped, but network hub regions based on Physarum centrality were contained local information as well as global information. It could reduce the in individual variation, and detect some regions which related to Post traumatic stress disorder, Schizotypal personality disorder and Alzheimer's disease. In this dissertation, I suggested some novel methodological frameworks for detecting network hubs and they could improve understandings of brain mechanism without problems in previous network hub studies.; 최근에 많은 연구들이 뇌의 기능이 복잡한 뇌 구조에서 어떻게 발현하고, 분석할 수 있는지에 대한 문제를 네트워크 관점에서 진행하고 있다. 그래프이론을 기반으로한 네트워크 분석을 하기위해 뇌의 각영역을 노드로, 그 연결성을 엣지로 정의한다. 네트워크의 군집정도와 평균거리등의 속성으로 뇌를 연구할 수 있다. 특히, 네트워크 허브는 네트워크 관점에서 가장 중요한 토폴로지 속성 중 하나로 간주되고 있다. 그러나 이전의 뇌 네트워크 연구에서는 네트워크 노드 해상도 선택 문제, 네트워크의 정보 흐름이 최단 연결을 통해서만 전파된다는 가정하에 분석이 진행된다는 문제등의 한계점들이 존재한다. 이 논문의 목적은 네트워크 허브 분석의 기존 문제점들을 보완 할 수 있는 새로운 방법들을 제안하는 것이다. 첫번째로, 뇌 네트워크 허브가 다양한 노드 해상도를 고려하여 결정되어야 한다는 가설을 기반으로, 다양한 범위의 노드 해상도에 걸친 허브 값의 평균에 의해 결정된 허브 강도를 사용하여 네트워크 허브를 정의하였다. 노드 해상도가 고려된 방법으로 나타난 전두엽의 일부 지역, 후두부 이랑 및 상두정소엽 이랑의 양쪽 대칭 영역이 다른 뇌 영역보다 허브 강도가 상대적으로 높았다. 또한, 이 영역들은 효율성에 대한 강도도 마찬가지로 다른 영역들보다 높은 경향을 보였다. 이 방법은 특정 네트워크 노드의 해상도의 결정없이 네트워크 허브를 찾아낼 수 있는 이점을 입증하였다. 둘째로, 네트워크의 정보 흐름이 가장 짧은 연결을 통해서만 전제된다는 기존 가정을 피하기 위해 피사럼 중심성 이론을 기반으로 뇌 네트워크의 허브 찾는 방법을 제안했다. 피사럼 모델을 이용하여 최단 경로 가정없이 관형 네트워크에서 유체 흐름 및 경로의 경쟁을 통해 관을 통하는 정보의 양을 제어함으로써, 두개의 공급원을 연결하는 최적의 경로를 찾을 수 있었다. 이 방법은 뇌 네트워크에서 허브를 찾기 위해 피사럼 모델을 사용한 첫 번째 연구였다. 뇌 네트워크에 피사럼 모델을 적용하기 위해 크게 다음과 같은 두가지의 과정이 필요하다. 1) 확산텐서이미징을 이용한 뇌 네트워크 구축, 2) 피사럼 모델을 뇌 네트워크에 적용후 피사럼 중심성 값을 계산. 기존에 뇌에 적용된 다양한 중심성 값과 비교해 봄으로써 피사럼 중심값의 성능을 분석하였다. 피사럼 중심성 이론을 기반으로 한 네트워크 허브 영역에는 글로벌 정보뿐만 아니라 로컬 정보가 포함된다는 결과를 볼 수 있었다. 또한, 피사럼 중심성 이론을 이용할 때, 그룹내에서의 개인적인 변이가 감소되는 경향을 보이고, 외상성 스트레스 장애, 조현병 및 알츠하이머 병과 관련된 일부 영역을 찾아 낼 수 있었다. 본 논문에서는 네트워크 허브를 생성하기 위한 몇 가지 새로운 방법론적 프레임 워크를 제안했으며, 이전 네트워크 허브 연구에서 제기되었던 한계점들을 보완해 뇌의 메커니즘에 대한 이해를 향상시킬 수 있음을 입증하였다.
