TY - THES AU - 박혜원 DA - 2010/08 PY - 2010 UR - https://repository.hanyang.ac.kr/handle/20.500.11754/141488 UR - http://hanyang.dcollection.net/common/orgView/200000415614 AB - 산업이 발달함에 따라 인간 생활의 편리함과 제품의 고급화를 가져다 주었지만 그와 더불어 환경오염이 수반되었다. 환경오염으로 인해 인간들은 환경 유해물질에 노출되고 이러한 영향으로 건강에 해를 입게 되었다. 이러한 이유로 환경 유해물질에 노출되었을 때 어떠한 유전적 변이가 있는지를 확인하고, 환경 유해물질의 위험성을 조기에 확인하고 분석할 수 있는 효과적인 분석 시스템이 필요하다고 생각된다. 현재 환경물질 위해성 예측을 위한 기술로 DNA chip 기술분야가 많은 발전을 이루고 있다. DNA chip은 그 종(species)에서 알려진 거의 모든 유전자들에 대한 발현 스크링이 가능할 정도로 기술이 발전하였으며, 미국 FDA에서는 이를 이용하여 환경 위해물질의 독성을 예측하는 기술을 개발하고자 많은 노력을 하였다. 여러 연구결과 전체의 유전자를 이용한 분석보다는 선별된 소수의 특정 유전자를 이용한 분석 방법의 정확도가 오히려 높다는 것을 알 수 있었다. 이러한 연구결과를 바탕으로 이 논문에서는 충분한 검증실험을 통해 선별 되어진 소수의 유전자를 이용하여 환경 유해물질의 독성을 평가 할 수 있는 DNA chip을 개발하고자 하였다. 이 논문에서는 환경유해물질에 특이적으로 반응하는 특정 유전자들을 선정하여 chip을 제작하였으며, 이를 HazChem Human Array라 명명하였다. 초기의 HazChem Human Array는 이전에 수행한 환경유해물질을 처리한 세포 시료로 수행한 Human whole genome chip을 이용한 microarray 실험 결과로 나온 유전자들 중 유의하게 발현변이가 있던300개의 유전자로 구성 되어지며, EDCs (Endocrine Disruptor Chemicals), PAHs (Poly cyclic aromatic hydrocarbons), VOCs (Volatile Organic Compounds) 그룹에 속하는 7개 환경유해물질을 이용하여 chip의 유용성을 확인하였다. 이후 Group classification의 신뢰도를 높이기 위해 유전자를 추가하여 600유전자를 포함하는 HazChem Human Array를 제작하였으며, EDCs, PAHs, POPs (Persistent Organic Pollutants), VOCs그룹에 속하는 13개 환경유해물질을 이용하여 chip의 유용성 및 신뢰성을 확인하였다. 최종적으로 여러 화학그룹에 속하는 많은 chemical들의 classification 및 독성을 판단하기 위해 유전자를 추가하여 1136개 유전자를 포함하는 HazChem Human Array 제작을 완성하였으며, EDCs, PAHs, POPs, VOCs, Pesticides 그룹에 속하는 17개 환경유해물질을 이용하여 chip의 유용성 및 신뢰성을 확인하였다. 실험결과 모든 chip 실험 시 각 환경유해물질 그룹에 특이적으로 발현하는 유전자들을 확인할 수 있었으며 이러한 유전자들의 발현 패턴 분석으로 실험에 사용된 환경유해물질이 어떤 유해물질그룹에 속하는지를 알 수 있었다. 위의 실험결과를 바탕으로, 개발된 HazChem Human Array를 이용하여 환경유해물질에 대한 유전자 발현 패턴을 분석할 수 있었고, 앞으로 이를 이용하여 많은 환경유해물질의 특성을 판단 할 수 있기 때문에, 빠른시간 내에 화학물질에 대한 특성과 정보를 얻을 수 있게 함으로써 toxicogenomics 연구분야에 많은 도움을 줄 것이라 생각된다.; Depending on the development of industry, it offer not only convenience of life but also side effects . In order to provide the necessaries, products were manufactured. For this reason, the environmental pollutions occurred and human was exposured to environmental hazardous chemicals. Environmental hazardous chemicals are present occurring at a very low levels within the human body, and are carcinogenic. Health risk valuation is a process by which the qualitative and quantitative effects that might occur when a person is exposed to environmental toxicities are estimated. Recently, a great deal of interest has been focused on the use of microarrays in toxicology to quickly classify toxicant based on characteristic expression profiles in a variety of species, and on the use of these profiles as a means for identifying the putative mechanism of action. Toxicogenomics using microarray technology offers the ability to conduct large-scale detections and those associated with alterations in mRNA stability or gene regulation. In this study, I developed the HazChem Human Array, which is expected to facilitate the identification of environmental toxicants. The early HazChem Human Array was composed 316 selected genes. In order to evaluate the viability of the manufactured early HazChem Human Array, I analyzed the gene expression profiles of 7 environmental toxicants; 2 PAHs chemicals, 2 POPs chemicals, and 3 VOCs chemicals. To improve the viability, secondary HazChem Human Array was composed 600 selected genes. In order to evaluate the viability of the manufactured secondary HazChem Human Array, I analyzed the gene expression profiles of 13 environmental toxicants; 2 EDCs chemicals, 3 PAHs chemicals, 2 POPs chemicals, and 6 VOCs chemicals. The final HazChem Human Array was manufactured using 1136 genes identified in previous studies. In order to evaluate the viability of the manufactured HazChem Human Array, I analyzed the gene expression profiles of 17 environmental toxicants; 2 EDCs chemicals, 3 PAHs chemicals, 2 POPs chemicals, 6 VOCs chemicals and 4 Pesticides. As a result, 17 environmental chemicals were separated into five chemical groups - EDCs, PAHs, POPs, VOCs and Pesticides. After the chip validations with 5 chemical groups, I conducted an expression profiling comparison of additional chemical groups using data analysis methods such as hierarchical clustering, 1-way ANOVA, SAM, and PCA. I selected 43 genes that evidenced significant alterations in expression with all 17 environmental toxicants. These results suggest that the HazChem Human Array will expedite the development of a screening system for environmentally hazardous chemicals at the level of toxicogenomics.
