dc.description.abstractStem cells have been considered as regenerative medicine for therapy of diseases. Especially, mesenchymal stem cells (MSCs) were known to have multipotent and were widely used for treating diseases. Recent studies have reported that hypoxia preconditioning enhanced the therapeutic potency of stem cells. However, teratoma formation, immune response and low viable rate were considered major disadvantages of stem cell-based therapies. Stem cell-derived exosomes, which played a role in cell-to-cell communication and were known as key regulators of the paracrine effect, were raised as a new alternative to overcome the limitations of stem cell-based therapies. In this study, we produced exosomes from human adipose-derived stem cells (HASCs) under hypoxic condition and used it for the treatment of pulmonary fibrosis. To prepare exosomes, tangential flow filtration (TFF) based multi-filtration system was used. We found that hypoxia-conditioned exosomes (Hy-EXOs) contained more anti-fibrotic factors and growth factors than normoxia-conditioned exosomes (Nor-EXOs). Hy-EXOs reduced expression of pro-fibrotic factors and attenuated pulmonary fibrosis. These results suggested that Hy-EXOs act as biochemical cue for regenerating lung tissue and provide a new approach to pulmonary fibrosis treatments.-
