Abstract
The three-dimensional (3D) printing system has been extensively utilized in the bone tissue engineering field by their precise functions to fabricate 3D scaffold for regeneration of the defected bone. The porous structure of the scaffold could help the new bone formation by promoting cell attachment, proliferation, and osteogenic differentiation. In this study, we fabricated the scaffold has the macro- and micro-porous features. The integrated techniques of the 3D printing and non-solvent induced phase separation (NIPS) were used to fabricate the macro-pore in the scaffold as well as the micro-pore on the strands of the scaffold. The micro-pores were successfully manufactured on the strands of the scaffold, which has similar mechanical properties of the cancellous bone. The human bone marrow stromal cells (hBMSC) was used to seed in the scaffold and analyzed in vitro to access the bio-functions of the scaffold. The cell attachment, proliferation, and differentiations in the highly porous scaffold were significantly increased than scaffold that has no pores on the strands of the scaffolds. These results suggest that the highly porous scaffolds has tremendous potential to use for regenerating the defected bone tissue.
