Abstract
마이크로 플루이딕스는 소형화된 유체 반응 시스템으로, 화학적, 생물학적 물질의 실시간 분석에 핵심기술로 여겨지고 있다. 다양한 구조를 갖는 마이크로 플루이딕스 기반 분석 시스템의 장점으로는 적은 양의 시료사용 및 폐기물 발생, 저비용, 빠른 시료분석, 휴대성, 일회성, 분석결과를 실시간으로 얻을 수있어 사용이 용이함을 들 수 있다. 이러한 분석 장치들은 특정 바이오물질을 검출하는데에 간편하고 유용하여, 자가진단 및 현장진단 (Point-of-care, POC) 분석장치로서 응용된다. 그리고, 마이크로 플루이딕스는 전기화학적 검출 방법과 결합하여 향상된 분석 성능을 보여준다. 전기화학적 검출 방법은 그 작동 방법이 간단하고, 휴대가 용이하며, 다양한 전극 제작이 가능하여, 각 공정과정을 간편하게 소형 센서에 집약할 수 있어 빠르고 저비용의 다양한 마이크로 플루이딕스 기반의 분석장치에 응용되고 있다. 또한, 나노입자의 넓은 표면적, 강한 촉매 효율, 독특한 광학적 특성, 높은 바이오물질과의 결합력과 전기화학적 촉매로서의 효율성으로 인해, 금속 나노입자를 증착한 센서는 그 향상된 분석 감도를 얻을 수있다. 본 연구에서 우리는 나노소재 기반의 마이크로 플루이딕 디바이스를 개발하였으며, 생화학분자를 전기화학적으로 검출하는데에 응용하였다. 먼저, 다공성 실리콘 기판에 전기화학적으로 형성된 금, 은 나노입자를 하이드록실아민의 정량 분석 및 표면 증강 라만 분광학 (surface enhanced Raman spectroscopy)을 이용한 로다민 6G 측정에 적용하였다. 또한, 단일벽 탄소나노 튜브 (SWCNT) 표면에 증착한 금-은 나노입자 (Au-AgNPs)를 마이크로 플루이딕 기반의 과산화 수소 센서에 전극으로 활용하였으며, 제작된 Au-AgNPs/SWCNT 전극의 전기화학적 반응속도를 연구하였다. 두가지 금속의 시너지 효과로 인하여 환원되는 과산화 수소를 효과적으로 검출하였다. 뿐만 아니라, 금, 백금 나노입자와 산화 그래핀 복합체 (Au@PtNPs/GO)를 비드 (Beads) 표면에 수식하여 인공 효소 유사체를 합성하였으며, 이 효소를 필름 기반의 마이크로 플루이딕 디바이스에 고정시켜 전기화학적인 과산화수소 검출 방법을 제시하였다. Au@PtNPs/GO 비드는 과산화효소 유사체로서 기질에 대한 높은 친화도를 갖으며, 효소반응에 의한 과산화 수소 측정에도 높은 감도를 보였다. 마지막으로, 측방 유동 정량법을 이용한 종이 기반의 스트립을 제작하여 당화알부민과 총 알부민을 정량 검출하였다. 보론산과 당화알부민의 Cis-diol 결합과 효과적인 발색 신호 측정을 위해 보론산이 수식된 아가로스 비드 (agarose beads)를 종이기반 스트립에 고정하였다. 제시된 검출방법은 낮은 검출한계를 갖으며, 넓은 농도범위에서 정량분석이 가능하다.; Microfluidics, the miniaturized reaction system to manipulate small volume of sample, has been considered as a key technology for the development of effective and real-time chemical and biological analysis. A number of microfluidic analysis systems have shown potential to meet requirements including the reduction of the total reagent and waste amount, low cost, fast sample analysis, portability, disposability and accessible to users by obtaining the detection results in real-time. This convenient sensing platforms provides an easy and useful way to detect specific biomolecules for self and on-site monitoring, referred to as Point-of-Care (POC) assay. Moreover, microfluidics takes the advantage of electrochemical detection to improve the performance of the miniaturized system. Electrochemical detection technique is widely applied in microfluidics due to their ease of operation, portability, simple modification process of electrode, and the ability to integrate multiple operation process on a single plaform, leading to time-saving and low cost system. The noble metal nanoparticles (NPs) modified onto the sensors enable to improve the sensitivity of biosensor due to its high surface area, strong catalytic efficiency, unique optical properties, the ability of covalent bonding with biomolcules and remarkable electrocatalytic activities. In this dissertation, we developed nanomaterials-based microfluidic devices and its application as electrochemical biosensors for biochemical molecules. First, electrochemically fabricated gold (Au) and silver (Ag) NPs on porous silicon substrate were applied for the quantitative analysis of hydroxylamine and the sensitive Surface-enhanced Raman spectroscopy (SERS) substrate for Rhodamine 6G. Moreover, we designed hydrogen peroxidase (H2O2) sensors with Au-AgNPs on single-walled carbon nanotube (SWCNT) microelectrode integrated in a PDMS microfluidic chip. The electrochemical kinetics of Au-AgNPs/SWCNT electrode in microfluidic chip was investigated. The Au-AgNPs bimetallic nanoparticles on SWCNT working electrodes was electrochemically deposited and then, enabled to synergize the electro-catalytic activity toward reduction of H2O2. Futhermore, the Au, platinum (Pt) NPs and graphene oxide (GO) hybrid structure immobilized film based electrochemical POC sensor was successfully developed for sensitive determination of H2O2. The Au@PtNPs/GO beads showed remarkable peroxidase-like activity with significantly stronger affinity toward 3,3',5,5'-tetramethylbenzidine (TMB) and H2O2. The on-chip electrochemical analysis of H2O2 using Au@PtNPs/GO beads showed a linear relationship corrensponding to H2O2 concentrations with high sensitivity. Finally, paper-based lateral flow assay (LFA) devices were produced for the sensitive detection of glycated albumin (GA) and total human serum albumin (tHSA). Boronic acid–derived agarose beads were packed into lateral flow channel to capture GA through specific cis-diol interactions and to enhance colorimetric signals by concentrating target molecules. The devices exhibited large dynamic ranges and low detection limits which covers the range of GA concentration in healthy plasma.
