dc.description.abstract | The main objective of this study was to develop aripiprazole-loaded solid self-nanoemulsifying drug delivery system to improve the aqueous solubility and dissolution rate by spray drying technique. Liquid solid self-nanoemulsifying drug delivery system (L-SNEDDS) was composed of oil (Captex 300) surfactant (Cremophor RH 60) and a co-surfactant (Transcutol HP) which were selected on the basis of their compatibility and solubility of aripiprazole (APZ). Pseudo-ternary phase diagram was constructed to detect the nanoemulsion region. The ratio of oil/surfactant/co-surfactant were selected in nanoemulsion region and APZ-loaded liquid SNEDDS (APZ-L-SNEDDS) was further optimized by particle size and polydispersity index. Spray drying technique was used to convert the optimized APZ-L-SNEDDS into APZ-S- SNEDDS with a suitable inert carrier such as silicon dioxide (SiO2) and calcium silicate (CaSiO3). APZ-S-SNEDDS were characterized by scanning electron microscopy, differential scanning calorimetry, powder X-ray diffractometry and in vitro dissolution study. APZ-L-SNEDDS was optimized with Captex 300/Cremophor RH60/Transcutol HP at the weight ratio of 20:35:45 using phase diagram. APZ-S-SNEDDS was successfully prepared by spray-drying method using 0.5 g of SiO2 and CaSiO3 as solid carriers. SEM, DSC and PXRD results suggested that APZ existed in amorphous form in APZ-S-SNEDDS. Moreover, APZ-S-SNEDDS significantly increased the aqueous solubility and dissolution rate of APZ. The developed solid self-nanoemulsifying system (S-SNEDDS) could provide a useful oral solid dosage form for poorly water-soluble drugs. | - |
