- The purpose of this study was to investigate the synergistic effects of prebiotics (grape seed flour) and probiotics (kefir-derived lactic acid bacteria) on liver steatosis in obese mice. High-fat diet (HFD)-induced obese mice were fed a high-fat (HF) diet supplemented with either grape seed flour (GSF), kefir-derived lactic acid bacteria (LAB), or both for 9 weeks. We observed a significant reduction in body and liver weights, plasma lipid concentration, and total liver lipid content in all test groups compared to the control group. In addition, the insulin resistance index was lowered in all test groups, indicating improved insulin metabolism. Supplementation with GSF and LAB together showed significant synergistic effects on weight gain, liver-function indicators, and short chain fatty acid content in the cecum. Microarray analysis of hepatic genes revealed additional significant changes in the expression of genes associated with liver function, metabolic disease, inflammatory response, and lipid metabolism. These results suggest that the synergistic effects of combined GSF and kefir-derived LAB on liver steatosis are mediated by modulation of the short chain fatty acid content in the cecum and expression of hepatic genes related to inflammatory metabolism and liver function.
