Abstract
현장진단은 환자 혹은 사용자의 혈액, 뇨, 타액 등의 체액 또는 의심이 되는 유해성분물질 등을 현장에서 수집하여 신속하게 분석하는 기술로, 전처리가 필요 없으며 수 분에서 1 시간 내에 분석 결과를 확인할 수 있다는 장점이 있다. 이러한 현장진단 기술은 임상현장에서의 진단을 목적으로 한 면역분석, 물, 식품, 유해물질 등의 환경분석, 폭발물, 마약 등의 화학/바이오 테러 분석 등에 폭넓게 활용되고 있다. 기존 실험실에서 대형장비를 이용한 분석법과 비교하여 현장검사는 비전문가가 짧은 시간 내에 분석할 수 있다는 장점이 있지만, 전반적으로 분석 민감도가 낮고 정확도가 높지 않아 분석기술의 응용이 제한적이다. 최근에는 현장진단의 한계인 민감도와 정확도를 높이기 위한 다양한 플랫폼 기술들이 개발되고 있다. 기존의 현장진단 기술이 갖는 문제점을 해결하기 위하여 본 연구에서는 다음과 같은 연구를 체계적으로 진행하였다. 첫째, 표적물질의 감도를 증진시킬 수 있는 신호측정 기술과 이를 정확히 해석할 수 있는 신호식별 기술을 개발하였다. 둘째, 미량의 표적물질에 대한 높은 민감도, 선택성, 재현성 등을 획기적으로 증진시킬 수 있는 광학 나노프로브를 합성하여 고감도 광학분석에 이용하였다. 셋째, 휴대성과 자동화 분석을 수행 할 수 있는 고감도 정량분석용 측방유동 디바이스를 개발하였다. 본 연구에서 이용한 라만분광법은 고체, 액체 등의 다양한 물질에 대한 정성/정량 분석이 가능한 광학분석 기술이다. 라만산란 신호는 표적물질의 분자진동 모드에 의존하기 때문에 물질에 따라 라만 신호가 다르게 나타나고, 특정 진동모드의 신호세기에 대한 변화를 추적하여 표적물질의 정량분석을 수행하는 것이 가능하다. 본 연구에서는 라만분광법, 나노입자 기술, 측방유동 디바이스 기술과 소형 분광기 등을 상호보완적으로 이용하여 폭발물과 유해물질에 대한 세가지 고감도 현장분석 기술을 개발하였다. 첫번째 연구에서는 14 종의 폭발물질에 대한 라만분광 신호를 측정하여 미지의 타겟 물질을 빠르게 확인할 수 있는 폭발물 판별용 라만분석 시스템을 개발하였다. 14 종의 폭발물질에 대한 라만분광 신호를 측정하여 이들에 대한 라이브러리를 구축하고 이를 고속으로 추적하여 판독할 수 있는 알고리즘을 개발하였다. 두번째 연구에서는 표면증강 라만산란 분광법을 이용한 고감도 폭발물질 검출 기술을 개발하였다. 표면증강 라만산란은 금속 표면의 전자기장 공명과 전하이동 효과에 의해 나타나며 라만신호를 1014~1015 배 증폭시킬 수 있다. 본 연구에서는 표면증강 라만 산란 현상을 유도할 수 있는 양극성 금 나노입자, 음극성 금 나노입자를 제조하고, 표적물질을 높은 선택성으로 검출할 수 있는 메이센헤이머 복합체를 이용하여 특정 폭발물질을 고감도, 고선택성으로 검출할 수 있는 분석기술을 개발하였다. 세번째 연구에서는 현장분석에 널리 이용되는 측방유동 면역분석 스트립에 표면증강 라만산란 분광법을 적용하여 고감도 정량분석, 빠른 검출시간을 유도할 수 있는 새로운 개념의 측방유동 디바이스를 개발하였다. 기존 측방유동 스트립은 신속성과 편이성 등 현장분석에 활용될 수 있는 많은 장점을 가지고 있지만 민감도가 낮고 정량분석이 불가능하다는 단점을 가진다. 본 연구에서는 이러한 단점을 극복하기 위하여 개량된 나노입자를 이용한 표면증강 라만산란 기반의 고감도 정량분석용 키트를 개발하고, 식중독균 외독소 단백질을 이용한 정량분석을 통해 성능을 검증하였다. 개발된 표면증강 라만산란 기반의 측방유동 키트는 육안으로 색변화를 판별하는 측방유동 키트나 효소면역분석법에 비하여 높은 감도와 넓은 정량분석 범위를 갖는 것을 확인하였다. 본 연구를 통하여, 현장진단 분석기술에 적용할 수 있는 라만분광법과 주성분분석을 이용한 분석물질 판별 라이브러리, 서로 다른 성질의 나노 입자를 이용한 표면증강 라만산란 기반의 고감도 분석, 기존 측방유동키트의 단점을 보완하는 표면증강 라만산란 기반의 고감도 정량분석용 측방유동키트 등이 성공적으로 개발되었다. 이러한 요소기술들은 현재 현장에서 이용되고 있는 분석기술의 낮은 민감도와 부정확한 판별을 보완하는 동시에 현장진단 분석기술의 응용범위를 한 단계 높일 수 있는 새로운 플랫폼으로 이용될 수 있을 것으로 기대된다.; Real-time methods are extensively used in the areas of environmental monitoring, chemical sensing and biological detection. Recently, these methods are applied to medical diagnostics of infectious diseases using the test from blood, saliva, urine or body fluids. These methods also provide a fast-analytical result in a few minutes to 1 hours. Real-time analytical methods have the advantage over conventional bench-top methods in terms of fast analysis time, low sample volume, simple interpretation. Nonetheless, their application has been still limited because of the lack of sensitivity and accuracy. Recently, many kinds of portable device have been developed and integrated with analytical devices to resolve the low sensitivity problem. In this dissertation, three different research projects, related with the real-time analysis, have been performed. First, we developed a fast Raman spectroscopic system for the identification of fourteen explosive materials have been developed. Their Raman spectral library has been constructed together with the fast recognition algorithm and portable Raman system for real applications in the field. Second, Raman detection nanoprobes for highly sensitive chemical detection has been fabricated. Surface-enhanced Raman scattering (SERS)-based detection system using nanoprobes has been applied to higly sensitive and reproducible detection of chemical materials. Third, a point-of-care device has been integrated with a portable detection system for the in-field analysis. A conceptually new device integrated SERS detection with lateral flow assay was developed for the highly sensitive and rapid detection to resolve existing problems associated with real-time analysis. Herein, we first report a rapid and sensitive recognition technique for explosive compounds using Raman spectroscopy and principal component analysis (PCA). Seven hundreds of Raman spectra for 14 selected explosives were collected, and were pretreated with noise suppression and baseline elimination methods. PCA, a well-known multivariate statistical method, was applied for the proper evaluation, feature extraction, and identification of measured spectra. Raman spectroscopy combined with PCA was found that three principal components achieved 99.3% classification rates in the collected spectra set. The results show that Raman spectroscopy in combination with PCA is well suited for the identification and differentiation of explosives in the field. We also report an ultra-sensitive SERS-based detection system for 2,4,6-trinitrotoluene (TNT) using nano-dumbbell structures formed by the electrostatic interaction between positively and negatively charged gold nanoparticles. Meisenheimer complexes were used to TNT specific binding methods between TNT and L-cysteine on gold substrates. As a result, many hot junctions were generated by the dumbbell structures from the electrostatic interaction between the negatively charged aromatic ring of the complex molecules and the positively charged nanoparticles, and the SERS signals were greatly enhanced. Our experimental results demonstrate that the SERS-based assay system using nano-dumbbells provides an ultra-sensitive approach for the detection of TNT explosives. It also shows strong potential for broad application in detecting various explosive materials used for real-time analysis. Finally, a novel SERS-based lateral flow immunoassay (LFA) biosensor was developed to resolve problems associated with conventional LFA strips (e.g., limits in quantitative analysis and low sensitivity). In our SERS-based biosensor, Raman reporter-labeled hollow gold nanospheres were used as SERS detection probes instead of gold nanoparticles. To verify the feasibility of the SERS-based LFA strip platform, an immunoassay of staphylococcal enterotoxin B was performed. The limit of detection for SEB, as determined with the SERS-based LFA strip, was estimated to be 1 pg/mL. This value is approximately three orders of magnitude more sensitive than that achieved with the corresponding ELISA-based method. The proposed SERS-based LFA strip sensor shows significant potential for the rapid and sensitive detection of target markers in a simplified manner.
