dc.description.abstract | 떡쑥(GnaphaliumaffineD.Don.)은두해살이초본식물로한국,중국,일본등아시아국가에서전통약재로사용되고있다.최근연구에서는항산화,항염증,항보체,항당뇨활성을갖는phenolics,flavonoids,그리고triterpenoids등의떡쑥화합물이연구되었다.이러한약리학적활성을갖는떡쑥은잠재적천연물소재의대체한약재로서식물에대한표준화가필요하다.하지만국산떡쑥의화학성분과검증된정량방법에대한품질관리기준에대한연구는아직보고되지않았다.따라서떡쑥지상부를분리,정제하여화학성분의구조동정후주요성분을동시에분석할수있는새로운분석법을개발하여떡쑥의식물화학성분에대한함량을측정하고자하였다. 본연구는떡쑥지상부로부터15개의phenolics(1–10,13–15,18,22),20개의flavonoids(12,16,17,19–21,23–25,27–37),2개의lactonecompounds(11,26)을포함하는37개의화합물을컬럼크로마토그래피법을이용하여분리하였고,화학적구조는NMR과HR-QTOF-ESI-MS를포함한분광학적자료를통해정확하게동정하였다.떡쑥의주요식물화학성분phenoliccompounds5,14,15,18과flavonoids16,17,19,20의8개성분을떡쑥의지표성분으로제안하고이들의동시분석법을개발하였다.확립된동시분석법에대하여ICH에서고시한의약품밸리데이션가이드라인을근거로하여분석법검증을수행하였다.설정된분석법은떡쑥의화학적프로파일링과정량화를위해주요성분분석을빠르고정확하게수행할수있다.이연구는국산떡쑥이함유된기능성식품또는천연물의약품개발에이용될수있을것으로사료된다.|GnaphaliumaffineisabiennialherbaceousplantthathasbeenusedasatraditionalmedicineinAsiancountries,includingKorea,China,andJapan.Inrecentstudies,thecontainsofG.affinearephenolics,flavonoids,andtriterpenoidsthatcontributedtothesepharmacologicalactivitiesincludingantioxidant,anti-inflammation,anti-complement,andantidiabetic.ThepharmacologicalactivitiesofG.affineareavalue-addedofnaturalsourcesasapotentialalternativeherbalmedicinethatthestandardizationmethodforthisplantisneeded.However,studiesonKoreanG.affine’schemicalconstituentsandqualitycontrolstandardofavalidatedmethodforquantitationhavenotyetbeenreported.Therefore,thepurposeofthisstudywastomeasurethecontentofphytochemicalcomponentsofG.affinebydevelopingananalyticalmethodthatcananalyzemajorcomponentsatthesametime,afterchemicalconstituentsisolatedandidentifiedfromtheaerialpartsofG.affine.Theseresultsshowedthecompounds1–37,15phenolics(1–10,13–15,18,22),20flavonoids(12,16,17,19–21,23–25,27–37),and2lactones(11,26),isolatedfromtheaerialpartsofG.affineusingchromatographicseparationsandtheirchemicalstructureswereaccuratelyidentifiedviaspectroscopicdata,includingNMRandHR-QTOF-ESI-MS.HPLCprofileanalysisforpatternrecognitionofG.affinewasestablishedusingtheverifiedeightphytochemicals,includingphenolics5,14,15,and18,andflavonoidcompounds16,17,19,and20,aremajorcomponentsofG.affine.Also,theHPLC-DADanalysismethodwasvalidatedforquantitationanalysisofthemainingredientsinG.affineaccordingtoICHguidelines.Theestablishedsimultaneousanalysismethodofmulti-componentsisrapidandaccurateforquantifyingthemaincomponentsofG.affineinadditiontothechemicalprofileofG.affine.Thisstudycouldbeusedtodevelopgoodqualityherbalproducts,includingfunctionalfoodsandhealthsupplementswithKoreanG.affine. | - |
