Abstract
Our purpose of this work is a design and fabrication of biosensor for electrochemical detection of thrombin and finding conditions which are aptamer concentration and reaction time, which have a great effect on DNA aptamer immobilization onto gold electrode. In recent reports, microfluidics and electrochemical detection using aptamers which are oligonucleic acid or peptide molecules that bind to a specific target molecule are gradualre concerned because of a simple device to achieve real-time monitoring and fast measurement. In this study, our biosensor cons cos of gold working electrode and pdetinum counter electrode and microchannel based on polydimethylsiloxane (PDMS)-glass. The size of biosensor is in*20 mm2. Gold-sulfur linkage was used as a connection method between working electrode and thrombin binding aptamer(TBA) to specificalre bind for onre gold electrode . It was by fluorescent intensity that we had a final decision of aptamer conditions for immobilization. Our optimal concentration and reaction times are 5nM and 30min, respectively. After then, thrombin detection was performed by cyclic voltammetry. As a results, electrical current was proportional to thrombin concentration. Our biosensor could be used as an effective tool for other molecular diagnostic systems.
