올레핀 링커에 의하여 형광염료가 표식된 유리딘 유도체의 합성

Abstract

현재까지 DNA 염기서열을 밝혀 내는데 사용되는 형광 표지 DNA (fluorescence-labeled DNA probe)을 이용한 Molecular beacon과 바이오센서(Bio sensor) 등에 대한 연구가 계속 되고 있다. 특히 SNPs(single nucleotide polymorphisms)와 돌연변이를 분석해 낼 수 있는 진단시약으로의 사용이 가능해졌다. 본 연구의 목적은 앞에서 언급한 5-(pyren-1-ylethynyl)-2’-deoxyuridine 를 기본 구조로 하여 비슷하게 디자인한 형광 뉴클레오사이드(Fluorescent nucleoside)에 대한 합성을 진행하는 것이다. |In the course of our work, we synthesized the olefin-linked fluorescent dye-labled 2‘-deoxyuridine derivatives by using the thymidine as a starting material. The oxidation of methyl group of thymidine, followed by the reaction with malonic acid in pyridine provided the 2‘-deoxyuridine containing the olefin-linker at 5-position. The fluorescein derivatives were used as the fluorescent dyes.
