- Antibiotics are useful for livestock to improve their living condition. But residual antibiotics induces some several human diseases in central nervous system, the heart, liver, skin, the gastrointestinal system, hearing and blood sugar metabolism, etc. Antibiotics have a different mechanism to kill some lives. To prevent these damage, it is necessary to design a sensitive detection system for early diagnosis of antibiotics. In this study, we developed a DNA aptamer specifically binds to benzylpenicillin. For screening benzylpenicillin-specific aptamer, we performed SELEX (Systematic evolution of ligands by exponential enrichment) rounds with reduced graphene oxide. Without any immobilization step, random ssDNA library (complexity; 1013 ~ 1018) incubated with reduced graphene oxide (rGO) for adsorption and eluted with benzylpenicillin. Through the 4 rounds of rGO-SELEX, we found a 60-mer aptamer sequence with a strong binding affinity (as 383.4 nM of Kd) to benzylpenicillin and low detection limit as ~ 5 nM. In addition, this aptamer showed specificity toward different kinds of antibiotics. Under these advantageous characteristics, we expect that this aptamer could be applicated to on-site detection system in groceries.
