Abstract
현미유는 현미에서 백미로 도정하는 과정에서 생기는 쌀겨(미강)에서 추출해 낸 기름으로 쌀겨에 함유된 감마오리자놀, 토코트리에놀, 천연 토코페롤 및 폴리코사놀 등 유용한 생리활성 성분들의 대한 기능성이 보고되고 있다. 또한, 현미유는 발연점이 높아 조리시에 유용하게 사용되며 맛이 고소하고 깔끔한 특성이 있어 식품가공에 적용 시 관능적 품질 향상에 도움이 될 수 있으나 유지의 특성상 수용성 제품에서는 별도의 유화공정을 거치지 않으면 저장안정성이 떨어지는 기술적인 어려움이 있다. 이에 현미유를 별도로 유화하여 저장안정성이 확보된 유화액 제조시 수용성 식품에 별도의 유화공정 없이 바로 적용가능한 산업적 이점이 있다. 본 연구의 목적은 반응표면분석법(response surface methodology)을 이용하여 현미유 유화액의 유화안정성에 영향을 주는 요인 변수들을 설정하고 요인변수와 유화안정성의 함수관계를 데이터로부터 추정하여 유화안정성을 최대화 할 수 있는 유화제조 공정을 최적화하는 것이다. 유화 제조 공정 최적화를 확립하기 위하여 반응표면분석법은 중심합성계획법에 따라 설계하였다. 독립변수는 유화제(폴리글리세린지방산에스테르, PGE)의 첨가량 (0.05-0.25%, X1), 호모믹서 회전속도(4,000-8,000 rpm, X2), 유상 대비 수상 비율(0.8-1.6, X3)로 설정하고 (-2, -1, 0, 1, 2)의 다섯 단계로 부호화(coding)하였고, 종속변수는 유화안정지수(emulsion stability index, ESI)로 하여 실험모델을 설정하고 적합성을 평가하였다. 각 독립변수에 따른 반응변수들의 분산분석 결과 유화제의 첨가량, 호모믹서 회전속도, 유상 대비 수상 비율에서 유의적인 이차 회귀식을 얻었고(p<0.05), 반응표면그래프와 등고선도를 통하여 유화 최적조건을 예측하였다. 반응표면분석법을 이용한 통계적 분석 결과치를 바탕으로 예측된 최적화 조건은 유화제 함량의 경우 현미유 유화액 총량의 0.2 %, 호모믹서 회전속도는 60˚C에서 6,700 rpm, 현미유와 물의 비율은 1:1.3 으로 분석되었다. 최적화 조건으로 제조한 현미유 유화물의 유화안정지수는 95.7 %로 반응표면분석모델의 예측치 94.4 %와 유사한 수준을 나타냈다. 반응표면분석법을 이용한 각 반응조건의 최적 조건에서 실제 실험을 수행하여 예측된 분산 안정성 확인 결과 transmission 및 backscattering 값이 시간경과에 따라 일치하는 것으로 나타났고, TSI (Turbiscan stability index)는 0.7 미만으로 나타나 응집이나 상층 부유 현상이 적고 분산성이 안정하게 유지되는 결과를 나타내 반응표면모델이 최적 유화물 제조 조건을 예측하는데 적합다고 판단하였다.; Brown rice oil is extracted from rice bran produced in the process of turning from brown rice to white rice and the useful physiologically active ingredients such as gamma orzanol, tocotrienol, natural tocopherol and polycosanol contained in rice bran have been reported. Because of its high fuming point, it is useful for cooking and good flavor. It can help improve taste quality when applied to food processing. However, due to the nature of the preservative, the products using water such as beverages are not utilized because of the technical difficulties of poor storage stability. The purpose of this study is to determine the optimum condition for rice bran oil emulsification using Response Surface Methodology(RSM). The RSM for establishing the optimization of the manufacturing process of emulsification is to add the amount of emulsifier (polyglycerin fatty acid ester, PGE) (0.05-0.25%, X1), the rotation speed of homomixer (4,000-8,000 rpm, X2) (2, -1, 0, 1, 2), the ratio of oil to water ratio (0.8-1.6, X3) and the dependent variable is the emulsion stability index(ESI) and the fitness was evaluated. As a result of analysis of variance of response variables according to each independent variable, emulsion stability index showed significant change according to amounts of emulsifier, the rotation speed of homomixer and the ratio of water to oil(p <0.05). Based on the results of the statistical analysis using the reaction surface methodology, the predicted optimization conditions were 0.2 g% of the total amount of the rice bran oil emulsion, 6,700 rpm of the homomixer rotation speed in 60℃ and the ratio of water to oil amounts is 1.3. The emulsion stability index of the rice bran oil emulsion prepared under the optimized conditions was 95.7%, which was similar to the predicted value of 94.4% of the RSM model. The transmission stability and the backscattering value were found to agree with each other over time and the turbiscan stablility index was less than 0.7, indicating that aggregation and upper floatation were less and dispersion stability was maintained .This optimum condition makes it possible to produce stable rice bran oil emulsion. It will increase the utilization of the rice bran oil in the food industry such as beverage production.
